Problems solved by technology

Chemical control measures are still effective measures to control RIFA, but they still cannot eradicate RIFA
Moreover, the long-term use of chemical agents to control RIFA will lead to unexpected rebound

Embodiment Construction

[0016] The present invention will be described in detail below in conjunction with the accompanying drawings.

[0017] 1 Materials and methods

[0018] 1.1 The source and rearing of tested insects

[0019] The RIFAs tested were collected from Wushan, Guangzhou, Guangdong. Referring to the method of Kuriachan et al., collect the RIFA colony from the field in a large plastic bucket and place it for 1-2 days. After the ant colony has established a new colony in the bucket, slowly drip tap water into the bucket to make the RIFA colony grow out of the colony. Move up and float out automatically. Use a 40-mesh net to transfer the ant colony into a plastic basin (7.0 L) coated with talcum powder at the mouth of the basin, and raise it indoors (room temperature 25°C, RH 65%-80%) with artificial feed for one week before testing. The criteria for judging worker ants, soldier ants, winged male ants, winged female ants, queens and larvae refer to the method of Zeng Ling et al. (2005). ...

Abstract

The invention discloses a method for controlling fire ants by using RNA interference and an application in reduction of the insecticide resistance of the fire ants. A dsRNA is designed and synthesized through dsCYP6Al-like and dsCYP4Cl-like CDs areas; the silence effects of a target gene and other homologous P450 genes are determined and the sensitivity of an exogenous compound is determined through a feeding method; and the detoxification metabolism action of the fire ants P450 on the exogenous compound is specified. The silence effects are detected through a qRT-PCR technology. The result shows that the silence effects of RNAi-mediated dsCYP6Al-like and dsCYP4Cl-like on dsCYP6Al-like and dsCYP4Cl-like mRNAs are significant after the dsRNA is fed for 24h; and compared with a control group, the silence efficiency reaches 64.6% and 77.1% separately.

Description

technical field [0001] The invention relates to the prevention and control technology of red imported fire ants, in particular to a method for preventing and treating red imported fire ants by using RNA interference and an application for reducing the drug resistance of red imported imported fire ants. Background technique [0002] RNA interference (RNA interference, RNAi) is a new method of gene regulation discovered in recent years (Matzke, 2001). That is, the use of exogenous or endogenous double-stranded RNA (dsRNA) specifically induces gene expression silencing. It uses RNA sequence matching to specifically identify target genes and inhibit target gene expression at the transcriptional level, post-transcriptional level or translational level, and has been widely used in gene function research, disease treatment and agricultural applications (Waterhouse, 2003; Matthew, 2004 ), at present, RNAi technology has been applied to insects of Lepidoptera, Orthoptera, Hymenopter...
