AI Technical Summary

Benefits of technology

This patented technology allows us to create an assay that uses two different types of particles called cholesteric acid crystal (CC) or polystyrene microspheres instead of traditional reagents like enzymatic substrates. These new materials have improved properties such as higher stability at room temperature compared to other chemical substances commonly found within them. They also provide better sensing capabilities than existing methods due to their ability to detect certain organisms without being affected by environmental factors. Additionally, they allow quicker analysis times while maintaining good accuracy when analyzed outside laboratories over long periods of time.

Problems solved by technology

The technical problem addressed in this patents relates to developing efficient ways to detect cryptoviruses like Cryptoparvoirus without being affected from other organisms present within their host range. Current testing procedures involve collecting samples containing blood drawn through tube feedings, making them time-consuming and expensive due to laborious sample collection process. Additionally, there may exist concerns over cross-contamination during analysis processes leading to false positives results.

Embodiment 1

[0031] Preparation of colloidal gold test strips for detection of Cryptosporidium parvum in cattle

[0032] 1. Method

[0033] (1) Firing of colloidal gold

[0034] Using trisodium citrate reduction method, to 99mL ddH 2 Add 1mL 1% chloroauric acid to O, add 1mL 1% trisodium citrate after boiling, until wine red, stop heating, and prepare 40nm colloidal gold particles.

[0035] (2) Determination of the optimal pH of the gold-labeled antibody

[0036] Use K 2 CO 3 Gradient labeling method, by observing the color change, determine the optimal pH of the gold standard.

[0037] (3) Determination of the optimal amount of antibody labeling

[0038] Use the antibody gradient labeling method to determine the optimal antibody labeling amount of the gold label by observing the color change.

[0039] (4) Preparation of gold-labeled antibody

[0040] Best K 2 CO 3 The amount of addition is to label the monoclonal antibody with colloidal gold, and the labeled gold-labeled antibod...

Embodiment 2

[0059] Colloidal gold immunochromatographic test strip performance test

[0060] 1. Method

[0061] In order to test the test performance of the test strip, a series of tests are required, including specificity tests, sensitivity tests, repeatability tests, and stability tests.

[0062] (1) Sensitivity test

[0063] Doubling dilution (1:1, 1:2, 1:4, 1:8, 1:16, 1:32, 1:64) of bovine Cryptosporidium parvum positive feces, observe the test strips according to the routine operation color situation. When the T line cannot be observed with the naked eye, and the C line is still clearly visible, the dilution at this time is the minimum detection concentration of the method.

[0064] (2) Specificity test

[0065] Take cattle Cryptosporidium parvum positive feces, negative feces, bovine giardia positive feces, and bovine Cryptosporidium anthrai positive feces as controls, and operate according to conventional methods to judge whether there is cross-reaction between various feces, s...

Abstract

The invention relates to double-antibody sandwiched colloidal gold immunochromatographic test paper for detecting zoonotic cryptosporidia. The double-antibody sandwiched colloidal gold immunochromatographic test paper prepared on the basis of CSpV viruses is used for detecting the zoonotic cryptosporidia; the specificity and the sensitivity are high, and by a double-antibody sandwiched method using a CSpV monoclonal antibody and a CSpV polyclonal antibody, the monoclonal antibody guarantees the specificity of the method and the polyclonal antibody improves the sensitivity of the double-antibody sandwiched colloidal gold immunochromatographic test paper, so that a detection result is more accurate. The prepared cryptosporidium detection colloidal gold test paper has the characteristics of rapidness, convenient operation, no need of a special instrument and the like; the detection result is clear and can be judged relatively quickly; and the double-antibody sandwiched colloidal gold immunochromatographic test paper is suitable for rapid diagnosis of a clinical site and large-scale epidemiological investigation in a pastoral area and the like.
