Variable domain library and uses

The invention provides polypeptides comprising a variant heavy chain variable framework domain (VFR). In some embodiments, the amino acids defining the VFR form a loop of an antigen binding pocket. In an embodiment, the polypeptide is a variable domain of a monobody and has a variant VFR. The polypeptide may optionally comprise one or more complementary determining regions (CDRs) of antibody variable domains. In an embodiment, the polypeptide is a variable domain of a monobody and has a variant VFR and one or more variant CDRs. Libraries of polypeptides that include a plurality of different antibody variable domains generated by creating diversity in a VFR, and optionally, one or more CDRs are provided and may be used as a source for identifying novel antigen binding polypeptides that can be used therapeutically or as reagents. The invention also provides fusion polypeptides, compositions, and methods for generating and using the polypeptides and libraries.

Inflatable device for use in surgical protocol relating to fixation of bone

A balloon for use in compressing cancellous bone and marrow (also known as medullary bone or trabecular bone). The balloon comprises an inflatable balloon body for insertion into said bone. The body has a shape and size to compress at least a portion of the cancellous bone to form a cavity in the cancellous bone and / or to restore the original position of the outer cortical bone, if fractured or collapsed. The balloon desirably incorporates restraints which inhibit the balloon from applying excessive pressure to various regions of the cortical bone. The wall or walls of the balloon are such that proper inflation of the balloon body is achieved to provide for optimum compression of the bone marrow. The balloon can be inserted quickly into a bone. The balloon can be made to have a suction catheter. The balloon can be used to form and / or enlarge a cavity or passage in a bone, especially in, but not limited to, vertebral bodies. Various additional embodiments facilitate directionally biasing the inflation of the balloon.

Expression of heterologous multi-domain proteins in yeast

This invention demonstrates the utility of a yeast expression system for the expression of functional heterologous multi-domain proteins in yeast. The yeast expression system allows for the inclusion of a plurality of (up to three) modular expression cassettes which may encode multiple polypeptide chains of a heterologous multi-domain protein on a single plasmid (Twin Cassette). Because multiple polypeptide chains may be encoded for by the expression cassettes of the present invention in a single vector, the system can produce equivalent amounts of the multiple polypeptide chains, thereby enhancing the yield of a functional heterologous multi-domain protein. For example, functional monoclonal antibodies (MAbs) comprising a heavy chain and a light chain of an immunoglobulin (IgG), and functional immunotoxins comprising an antibody domain and an oxidase toxin may be produced using the Yeast expression system of the present invention. In addition, functional single chain antibodies, antibody fragments and chimeric antibodies may also be produced.

Rear-mounted aerodynamic structure for truck cargo bodies

ActiveUS20080309122A1Reduce resistanceRapid and easy transitionVehicle seatsWindowsEngineeringActuator
This invention provides an aerodynamic structure attached to the rear face of a truck cargo body, which rear typically contains a door assembly, with a plurality of doors that swing open on hinges, or a single, full-width door, which rolls upwardly. The various embodiments of the invention allow an aerodynamic structure to be permanently attached to the rear of the trailer in a manner that would obscure access to the door(s) in a deployed position, in which the aerodynamic structure generates reduced drag on the trailer body, yet enables ready access to the door(s) in a folded position. The folded position still allows the rear of the trailer to be fully accessible for loading and unloading, and in the case of swinging, hinged doors (among others), allows the doors to be folded through a full 270-degree arc from a closed position to a position flush along the sides of the vehicle, with a minimal sideways projection. The various embodiments also enable relatively rapid and easy transition between the folded position and the deployed position using a variety of actuators and linkages that tie the folding and deployment of various panels of the structure together. This allows an operator to selectively fold and deploy the structure without undue effort or strength.

Collapsible container

A collapsible container has four panels that fold down on a base for storage. The base has slide sockets with vertical slots. Front and rear panels pivot on the base with slide hinges received in the slide sockets. The panels are able to slide up and down. A hinge pin has a narrowed waist portion to engage a latch. Left and right panels pivot with snap hinges received in snap sockets on the base. Trunnions, disposed in the snap sockets, are eccentric to increase or decrease the backlash between the snap hinge and the snap socket as the panels pivot. The left and right panels have edge pockets that receive edge fingers on the front and rear panels to hold the container open. The front and rear panels each have an access opening with access doors. Links pivot the access doors on two hinge rods. Locking members wedge the hinge rods in the hinge knuckles. The base has feet with bumper plates having flanges that prevent the container from shifting on an adjacent lower stacked container.

Sanitary napkin with compressed grooves

A sanitary napkin in which diffusing liquid hardly reaches a rear end edge is adapted to make a liquid absorbent layer less deformable in a rear region so that it can easily be kept in close contact with the wearer's buttocks. The sanitary napkin has an elongated main absorbent region surrounded by compressed grooves that are formed in a skin-side surface. Since a rear end of the main absorbent region is far away from a rear end edge of the napkin to provide a relatively large rear region, worries about rearward leakage may be relieved. Furthermore, since auxiliary longitudinal grooves are provided in the rear region, a liquid absorbent layer may hardly be twisted.
