[0070] In one embodiment, an injectable thermosensitive composite hydrogel is employed, wherein the hydrogel is liquid during injection and then gels (gels at human body temperature) while in vivo. In one embodiment, injectable thermosensitive composite hydrogels (such as hydrogels with hyaluronic acid, such as chemically cross-linked and high molecular weight Gel One, and F127) are used to deliver therapeutic agents, for example say, loading of amobarbital on the hydrogel reversibly inhibits the respiratory enzyme complex I, a key mediator of chondrocyte damage after shock. The hydrogel hardens after injection (prevents leakage from the joint), enabling the therapeutic agent to remain in the joint area, eg, for about 3 days after injection into the joint injury site. In one embodiment, the hydrogel comprises 17% (w / v) F-127 and 0.2% (w / v) HA and is loaded with 2.5 mM amobarbital. At the injury site, a thermosensitive hydrogel anchors amobarbital, preventing chondrocyte death...

Abstract

The invention provides compositions. The compositions comprise a reverse-temperature sensitive hydrogel comprising a biopolymer such as a polysaccharide and a synthetic polymer, and a compound in an amount that reversibly inhibits respiratory enzyme complex I, and methods of using the composition, are provided.

Description

[0001] priority claim [0002] This application claims priority to US Provisional Patent Application No. 62 / 207,059, filed August 19, 2015, the disclosure of which is incorporated herein by reference. [0003] Statement of Government Rights [0004] This invention was made with Government support under Grant No. W81XWH-11-1-0583 awarded by the Department of Defense. The government has certain rights in this invention. Background technique [0005] Most older adults are familiar with the pain, immobility, and general disability associated with osteoarthritis. Post-traumatic osteoarthritis (PTOA) is an acutely accelerated form of arthritis associated with traumatic damage to the articular surfaces of a joint, resulting in a well-received joint replacement procedure considered common in orthopedic medicine in patients. Candidates had previously caused substantial disease progression. Because patients are often injured at a relatively young age and there are currently no vi...
