Problems solved by technology

[0003] Previous experiments proved that MaPIP1; 1 transgenic Arabidopsis can respond to drought stress (Xu et al.2014), however element analysis did not find any known cis-acting elements induced by drought or salt stress in the 1362bp promoter element

Embodiment Construction

[0017] The technical solutions in the embodiments of the present invention will be clearly and completely described below with reference to the accompanying drawings in the embodiments of the present invention. Obviously, the described embodiments are only a part of the embodiments of the present invention, but not all of the embodiments.

[0018] refer to figure 1 , a screening method for the core region of a banana aquaporin gene promoter, comprising the following steps:

[0019] S1: control selection, with transgenic Arabidopsis thaliana transformed with the CaMV35S promoter as a control;

[0020] S2: Detection of GUS activity of promoter, detection of GUS activity of promoter was performed on leaves and roots of transgenic Arabidopsis thaliana seedlings treated with 100mM, 200mM and 300mM 18% PEG6000 for 15 days;

[0021] S3: Observe the staining situation. From the phenotype point of view, in GUS staining, except for the CaMV35S control plant, the leaves of the promoter ...

Abstract

The invention discloses a screening method of a banana water channel protein gene promoter core area. The screening method comprises the steps as follows: control selection: transgenic arabidopsis forconverting a CaMV35S promoter as a control; detection of promoter GUS activity: promoter GUS activity detection is performed on leaves and root systems of 15-day transgenic Arabidopsis seedlings treated by 100 mM, 200 mM and 300 mM 18% PEG 6000; and observation of staining condition: seen from phenotype, in GUS staining, except CaMV35S control plants, promoter transgenic plant leaves of each fragment is not stained or lightly stained while root tip parts are deeply stained in transgenic root systems. According to the result, the core area of MaPIP1:1 promoter is recognized, and the acting mechanism of the MaPIP1;1 gene during resisting of drought stress can be further illuminated.

Description

technical field [0001] The invention relates to the field of biotechnology, in particular to a screening method for the core region of a banana aquaporin gene promoter. Background technique [0002] In the early stage of banana, the plant is small, the root system is shallow, and it is susceptible to drought. Its leaf area is large and the amount of transpiration is large, especially in the high temperature season. The earth damages the normal physiological and metabolic activities of bananas, resulting in reduced production and quality of bananas. In banana cultivation and production, water consumption is large. For every 500 grams of dry matter produced, 300 kilograms of water needs to be absorbed. In the vegetative growth stage of bananas, drought will cause poor growth and development of banana vegetative organs, and the growth rate and amount of growth will be significantly reduced. When the drought is severe, the leaves will droop, wither and wither, the stomata will ...
