Abstract

The invention discloses a method for efficiently promoting germination of premnapuberula seeds. The method comprises the following steps: cleaning mature premnapuberula fruits into clean water, removing peel, performing cleaning, and selecting clean seeds for later use; soaking the seeds for later use into a sulfuric acid solution for 3-8 minutes, cleaning the seeds completely with clean water, and sucking off moisture on surfaces of the seeds by using filtering paper for later use; soaking the seeds for later use into a gibberellin solution, taking out the seeds, completely flushing the seedswith clean water, and sucking off moisture on surfaces of the seeds by using filtering paper for later use; flushing the seeds for later use by using 75% alcohol, 0.1% mercury bichloride and sterilewater for multiple times to implement sterilization treatment so as to obtain sterilized seeds for later use; and inoculating the sterilized seeds with an MS (Murashige&Skoog) culture medium, and performing germination culture. By adopting the method, germination difficulties caused by physical dormancy of the premnapuberula seeds can be effectively solved, the germination rate of the premnapuberula seeds can be increased, the germination time can be shortened, then support is provided for on-scale production of the premnapuberula seeds, and in addition, beneficial effects of simplicity in operation and low cost can be achieved.
