Abstract

The invention discloses a sudden cardiac death susceptibility detection kit based on STIM1 gene insertion and deletion polymorphic sites. A specific primer pair contained in the kit is designed for aninsertion and deletion site No. rs3061890 on an STIM1 gene, DNA fragments containing the site can be specifically amplified, and different genotypes are recognized by detecting the mobility of the fragments with different lengths in capillary electrophoresis. It is found that in combination with our case control study, it can be considered that a person whose insertion and deletion site No. rs3061890 on the STIM1 gene of a detected DNA carries a deletion type allele is of an SCD susceptible type. Therefore, the technology can play a role in predicting the SCD susceptibility of an individual by detecting the genotype of the insertion and deletion site No. rs3061890 on the STIM1 gene of the individual; the subject group proves for the first time that a tetrabasic group (TCTC) insertion anddeletion polymorphism (rs3061890) in 3'UTR of the STIM1 gene is significantly associated with the risk of suffering from SCD; and the frequency distribution of the polymorphism in the Asian populationis that an insertion type is 0.669, and a deletion type is 0.331.
