Embodiment 1

[0010] 120 healthy and clean SD rats were randomly divided into sham operation group (group A), model group (group B) and hyperbaric oxygen group (group C), with 40 rats in each group. 8 rats were randomly selected from each group on 4d, 7d, 14d, 21d, and 28d after operation for various tests, 5 rats in each group were subjected to immunohistochemical experiments; 3 rats in each group were subjected to fluorescent quantitative PCR experiments.

[0011] After intraperitoneal anesthesia with 10% chloral hydrate, the rats were fixed on the rat brain stereotaxic instrument, and the model was made by local disinfection according to the principle of aseptic operation;

[0012] The rats in the hyperbaric oxygen group were placed in the animal cabin 3 hours after the model was made, the cabin door was closed tightly, and the oxygen flow rate was uniformly variable at 5-8 L / min to pressurize for 15 minutes. When the pressure in the cabin reached 0.20 MPa, it started Stabilize the press...

Abstract

The invention discloses a method for researching angiogenesis influence in the brains of rats suffering from intracerebral hemorrhage caused by hyperbaric oxygen. SD (Sprague Dawley) rats randomly comprise a sham-operation group (group A), a model group (group B) and a hyperbaric oxygen group (group C). The neovascularization condition of brain tissues around hematoma of rats in each group is observed under an HE dyeing microscope by adopting a tissue slice; the protein expression content of HIF1 (Hypoxia Inducible Factor)-alpha and VEGF (Vascular Endothelial cell Growth Factor) in the brains of the rats in each group are detected by adopting an immunohistochemistry analysis method; the expression level of the HIF1-alpha mRNA and VEGFmRNA of the brain tissues of the brains in each group is determined by adopting a fluorescent quantitation RT-PCR (Reverse Transcription-Polymerase Chain Reaction) method.

Description

technical field [0001] The invention relates to a test method, in particular to a method for studying the effect of hyperbaric oxygen on neovascularization in the brain of rats with cerebral hemorrhage. Background technique [0002] Neovascularization after intracerebral hemorrhage (ICH) increases blood and oxygen supply to damaged brain tissue, which is beneficial to brain tissue repair. Hyperbaric oxygen (hyperbaricoxygen, HBO) can increase the oxygen supply of brain tissue, improve microcirculation, promote the regeneration of blood vessels in damaged tissues, and promote the recovery of nerve function. However, there are few domestic and foreign studies on the mechanism of hyperbaric oxygen on angiogenesis in cerebral hemorrhage, and the mechanism is still unclear. Contents of the invention [0003] After intraperitoneal anesthesia with 10% chloral hydrate, the rats were fixed on the rat brain stereotaxic instrument, and the model was made by local disinfection accord...
