Abstract

The method comprises the steps that fresh and tender stem segments with buds are obtained from aseptic seedlings and processed and grafted to an initial culture medium for culture, and detoxified stem segments with buds are obtained; multiplication culture is performed, wherein the detoxified stem segments with buds are grafted to a multiplication culture medium containing MS, 1.25 mg/L of 6-BA, 0.25 mg/L of NAA, 0.25mg/L of IBA, 30g/L of sucrose and 6.5-7g/L of agar powder, and culture is performed to obtain sub-cultured seedlings; rooting culture is performed, wherein the sub-cultured seedlings are transferred to a rooting culture medium containing MS, 1.25 mg/L of IBA, 0.05mg/L of NAA, 30g/L of sucrose and 6.5-7g/L of agar, and culture is performed to obtain rooting seedlings; the seedlings are hardened and transplanted, wherein the rooting seedlings are transferred to a greenhouse for seedling hardening, and transplantation is performed after seedling hardening. By adopting the method, the Beijing mountain ash multiplication efficiency can be improved to 26.5, the rooting rate can be up to 93.3%, the survival rate of the hardened seedlings is 100%, and the method is conductive to quantified production of Beijing mountain ash.
