Embryo treatment method and special medium for improving adventitious bud induction rate of mature embryo cotyledon of plum blossom

A treatment method and mature embryo technology, applied in the field of plant tissue culture, can solve the problems of reducing the induction rate of adventitious buds, the low degree of differentiation of cotyledons near the hypocotyl end, and the low transformation efficiency of genetic transformation systems, achieving shortened induction time and effective Good for seedling growth and improved regeneration efficiency

Problems solved by technology

[0004] The mature embryo cotyledons of plum blossoms treated by the above method can completely remove the radicle, hypocotyl, and germ of the seed, and there will be no germination phenomenon in the later culture, but this embryo treatment method will differentiate the cotyledon near the hypocotyl end. A part of the cotyledon with low degree and high regenerative ability is also removed, which greatly reduces the induction rate of adventitious buds
According to the test statistics, the average regeneration rate of mature embryo cotyledons of plum blossoms treated with the above-mentioned embryo treatment method is generally about 12.50% to 30.15%, and the lower regeneration rate leads to lower transformation efficiency of the genetic transformation system of this regeneration system.

Embodiment 1

[0039] Example 1 Test material selection, culture medium design and inoculation culture

[0040] 1. Test materials and treatment:

[0041] The test materials of the present invention are four strains 6-5, 11-1, 12-1, 14-2 of the plum blossom variety 'Xuemei' × 'Fenpi Gongfen' hybrids (the implementation of the present invention is not limited to the above four Strains, the implementation of the present invention and its effects have no restrictions on plum blossom genotypes and varieties, and the mature embryo cotyledons of strains) are test materials, and the collection site is the plum blossom cultivation base in Huazhong Agricultural University. The collection time is the first ten days of June (that is, the fruit about 110 days after the plum blossoms bloom). The collection standard is that the fruit is fully mature and falls off naturally, free from diseases and insect pests, the pulp is removed within one week after harvest, the core is kept, dried in the sun and stored...

Embodiment 2

[0057] Example 2 Effects of different embryo treatment methods on the regeneration rate of cotyledons of mature plum blossom embryos

[0058] Embryo treatment had a significant effect on the regeneration of cotyledons from mature embryos of plum blossoms. The mature embryo cotyledon of 4 strains (through embodiment 1) that embryo treatment mode of the present invention (pick out germ, hypocotyl, radicle) process, its adventitious bud induction rate is all significantly higher than original embryo treatment mode (excise germ, hypocotyl, radicle) treated mature embryonic cotyledons (Table 3).

Abstract

The invention belongs to the field of plant tissue culture, and relates to an embryo treatment method for increasing the adventitious bud induction rate of cotyledons of mature embryos of plum blossoms, and a special culture medium. The method comprises the steps of (a) cutting off cotyledons, far away from hypocotyl ends, of mature embryos of disinfected plum blossoms, and reserving cotyledons close to the hypocotyl ends; (b) peeling seed coat with a scalpel, inserting the tip of the scalpel into a gap between the every two cotyledons, and separating the two cotyledons; (c) gently pricking and removing radicles, hypocotyls and germs at the top ends of the inner sides of the cotyledons with the tip of the scalpel, enabling all the radicles, hypocotyls and germs to be thoroughly removed, and reserving the cotyledons near the hypocotyl ends as much as possible; (d) after the radicles, the hypocotyls and the germs are removed, cutting each cotyledon of the mature embryos into two parts along the middle line so as to obtain four cotyledon explants, close to the hypocotyl ends, of the mature embryos; (e) inoculating the cotyledons, close to the hypocotyl ends, of the mature embryos intoa regeneration medium, inducing adventitious buds, enabling the adventitious buds to elongate and carrying out rooting culture so as to obtain regenerated plants. The invention also discloses the special culture medium. After the embryo treatment method provided by the invention is adopted, the adventitious bud induction rate of the cotyledons of the mature embryos reaches 42.64-50%.

Description

technical field [0001] The invention belongs to the technical field of plant tissue culture and is related to garden plant tissue regeneration, in particular to an embryo treatment method and a special culture medium for improving the adventitious bud induction rate of mature embryo cotyledons of plum blossoms. The present invention is a new type of embryo treatment method used in the regeneration and cultivation of mature embryo cotyledons of plum blossoms. The induction rate of adventitious buds of mature embryo cotyledons of plum blossoms treated by this new embryo treatment method is significantly higher than that of the original embryo treatment method. Ripe cotyledons of plum blossoms. The invention also relates to a special culture medium. Background technique [0002] Plum blossom (Prunus mume Sieb.et Zucc.) belongs to Rosaceae (Rosaceae) Prunus (Prunus) plants, small trees, flowering winter and spring. In Chinese garden and flower culture, plum blossom has a pivot...
