Embodiment 1

[0019] 1 Materials and methods:

[0020] 1.1 Experimental materials:

[0021] In Yusheng Cashmere Goat Breeding Company in Huan County, Gansu Province, 11 Longdong Cashmere Goat rams were used as male parents, and Longdong Cashmere Goat ewes were artificially inseminated. Among the offspring produced, 373 Longdong cashmere goats were randomly selected as the test subjects. When the offspring reached one year old, the cashmere production was measured on the spot, and the cashmere height was measured at the midline of the back. At the same time, cashmere samples were collected at the midline of the back for laboratory determination of cashmere fineness. In addition, 8 ml of blood was collected from the jugular vein of each sheep, anticoagulated with citric acid dextrose (ACD), and frozen at -20°C for extraction of genomic DNA.

[0022] 1.2 Genomic DNA extraction:

[0023] Blood genomic DNA was extracted by conventional phenol-chloroform method.

Abstract

The invention discloses a genetic marker related to a cashmere wool property of eastern Gansu cashmere goats. The genetic marker is a partial DNA (Deoxyribonucleic Acid) sequence of a goat KRTAP20-2 gene; a nucleotide sequence of the genetic marker is shown as SEQ ID No: 1 to SEQ ID No: 3 in a sequence table; the invention provides an amplification primer and application of the genetic marker. Thegenetic marker disclosed by the invention has the beneficial effects that a KRTAP20-2 gene type and an allele have extremely remarkable or remarkable influences on the wool yield and the wool layer height of the eastern Gansu cashmere goats. The wool yield of individuals containing an allele A is 38g higher than that of individuals without the allele A; the wool yield and the wool layer height ofindividuals containing an allele B are 30g and 0.1cm lower than those of individuals without the allele B respectively. Therefore, when the wool yield of the eastern Gansu cashmere goats is subjectedto breed selection, the individuals containing the allele A need to be remained, and the individuals containing the allele B are weeded out, so that the aim of improving the wool yield of the easternGansu cashmere goats is realized.

Description

technical field [0001] The invention relates to a molecular marker breeding technology related to cashmere traits of Longdong cashmere goats, in particular to a molecular marker related to cashmere traits of Longdong cashmere goats and an application thereof. Background technique [0002] Cashmere refers to the hair fiber with a diameter of 25um or less in the goat coat. It is an unmyelinated hair fiber formed by the secondary hair follicle in the goat skin. It is called "Cashmere" in the international market. Because of its many advantages such as fineness and softness, good luster, and strong warmth retention performance, cashmere is expensive and is called "soft gold". Gansu Province is one of the main cashmere producing areas in China. There are two unique cashmere goat breeds in Gansu Province, the Hexi Cashmere Goat and the Longdong Cashmere Goat. Among all cashmere traits (such as cashmere yield, cashmere layer height, fineness, net cashmere rate, etc.), cashmere yie...
