Embodiment 1

[0041] Selected clinical samples from Lanxi Red Cross Hospital, the Second Affiliated Hospital of Xi’an Jiaotong University, Xijing Hospital, and Tangdu Hospital from 2016 to 2017 with colonoscopy case results, including 77 cases of colorectal tumors and 204 cases of asymptomatic people Stool samples. The above-mentioned samples were subjected to gene capture and high-throughput sequencing, and their clinical and pathological information were collected at the same time. Finally, the genome sequencing results and clinical information were comprehensively analyzed to construct a colorectal tumor cell detection model.

[0042] (1) Gene capture and high-throughput sequencing

[0043] Perform gene capture and high-throughput sequencing of stool samples in our laboratory (see figure 1 ). Extract the genomic DNA of the sample to be tested, use the probe to capture the target genes KRAS, NDRG4; use the bisulfite conversion method to transform the NDRG4 gene to retain the methylated C base...

Embodiment 2

[0065] List 6 volunteers who went to Lanxi Red Cross Hospital, the Second Affiliated Hospital of Xi’an Jiaotong University, Xijing Hospital, and Tangdu Hospital to use the variable results measured by NGS technology to obtain the consistency judgment process using this model.

Abstract

The invention provides a method and a system for determining colorectal tumor cells. The method comprises the following steps: obtaining a KRAS gene and an NDRG4 gene of a sample to be tested respectively, and methylating the NDRG4 gene; sequencing the KRAS gene and the methylated NDRG4 gene respectively to obtain a KRAS gene sequencing result and an NDRG4 gene sequencing result; performing firstcomparison on a gene sequence at a first target position in the KRAS gene sequencing result and a first reference sequence, and calculating the mutation rate of the KRAS gene; performing second comparison on a gene sequence at a second target position in the NDRG4 gene sequencing result and a second reference sequence, and calculating the methylation linkage ratio of the NDRG4 gene; based on the mutation rate of the KRAS gene and the methylation linkage ratio of the NDRG4 gene, determining whether the sample to be tested has the colorectal tumor cells. Through the method and the system, whether colorectal tumor cells exist can be quickly, simply and specifically detected, and the sensitivity is high.

Description

Technical field [0001] The invention relates to the field of molecular biology technology. Specifically, a stool DNA colorectal tumor multi-gene prediction model based on NGS technology. More specifically, the present invention relates to a method and system for identifying colorectal tumor cells based on KRAS and NDRG4 genes. Background technique [0002] Colorectal cancer (Colorectal cancer, CRC) is a common malignant tumor, including colon tumors and rectal tumors, also known as colorectal tumors, which refer to malignant tumors that occur in the lower digestive tract, colon or rectum, and are one of the main cancers threatening human health. First, its incidence rate ranks third among male tumors and second among female tumors globally. In my country, with the development of society and economy, changes in residents' living habits, dietary structure, etc., the incidence and mortality of colorectal tumors in my country continue to rise. China's cancer statistics in 2015 sho...
