Problems solved by technology

So far, studies have confirmed that serotonin receptor 4 (HTR4) can significantly affect the gastric emptying and feeding ability of animals. It has been reported in human and mouse gastric motility studies, and also in rainbow trout and Schizothorax Research on serotonin antagonists, but there is no report on the application of SNPs related to HTR4 in puffer puffer

Embodiment Construction

[0028] The technology described in the present invention is further described in detail through specific examples.

[0029] The present invention is based on a pair of primers described in SEQ ID NO: 2 and SEQ ID NO: 3 to extract Fugu obscura genome DNA, perform PCR amplification, sequence the amplified product and analyze the sequencing result. The SNP loci (SNPg.100C>G) significantly correlated with the fatness of Puffer obscurus were obtained, which can be applied to molecular marker-assisted breeding of Puffer obscurus.

[0030] Below will further illustrate by specific embodiment

[0031] a) The acquisition of puffer puffer populations;

[0032] b) Extraction of Fugu obscura genome DNA;

[0033] c) based on the SNP primers, PCR amplification is carried out to the genomic DNA of puffer pufferfish;

[0034] d) Sequencing the PCR amplification product, and determining the nucleotide type of the SNP based on the sequencing result;

[0035] e) SNP site nucleotide type and ...

Abstract

The invention discloses an SNP molecular marker related to the condition factor of Takifugu fasciatus and applications thereof. The gene sequence of SNP molecular marker source Takifugu fasciatus serotonin receptor 4 (HTR4) is as shown in SEQ ID NO:1, an SNP site exists at the 100th position of the sequence, and the base is C or G. The SNP locus disclosed by the invention is obviously related to the condition factor of the Takifugu fasciatus, and can be effectively used for molecular marker-assisted breeding of the Takifugu fasciatus, so that the breeding time of improved fish varieties can besaved. The method is accurate in operation, simple and practical.

Description

technical field [0001] The invention relates to a SNP molecular marker related to the fatness of puffer puffer obscurus and its application, and belongs to the technical field of aquatic animal genetics and molecular marker-assisted selection breeding. Background technique [0002] Takifugu fasciatus (Takifugu fasciatus), commonly known as river fugu, belongs to the class Osteophytes, subclass Radix, Fugiformes, and the genus Fugu. It is a delicious and valuable economic fish. Known as "three delicacies of the Yangtze River", they are mainly distributed in coastal areas and inland rivers of countries such as China, the Korean Peninsula and Japan. Until 1954, the output of wild puffer fish in the lower reaches of the Yangtze River could reach 1,000 tons per year. However, with the rapid development of fishing technology for half a century, uncontrolled fishing has caused the population of puffer puffer to decline rapidly, resulting in the fishing season of puffer puffer obscu...
