Embodiment 1

[0026] Example 1: Hydrolysis at acidic pH

[0027] According to US Patent 6,005,073 (pH 2.8; 10,000 g force), a chicken protein isolate derived from myofibril protein and sarcoplasmic protein was produced from fresh chicken breast muscle and adjusted to pH 5.5 to precipitate the protein. Then hydrochloric acid (2N) was added dropwise to adjust the precipitated protein back to pH 3.5. Except for the acid, no other additives are added. The protein concentration of the acidified protein was 49.86 mg / ml. Put two aliquots of the protein sample into a glass beaker and put it in a 50°C water bath. Mix with a spatula in one of the beakers to disperse 0.05% (w / w) S-16774 Enzeco Fungal Acid Protease (Enzyme Development Corporation, 21 PennPlaza, 360 West 31 st St., New York, NY). The samples were incubated at 50°C for 2.3 hours. After that, sodium hydroxide (2N) was added dropwise to adjust both samples to pH 7.0. The sample without enzyme precipitated at pH 5.5, but returned to a liquid ap...

Embodiment 2

[0035] Example 2: Hydrolysis at neutral pH

[0036] According to US Patent 6,005,073 (pH 2.8; 10,000 g force), a chicken protein isolate derived from myofibril protein and sarcoplasmic protein was produced from fresh chicken breast muscle and adjusted to pH 5.5 to precipitate the protein. Then sodium hydroxide (2N) was added dropwise to adjust the precipitated protein to pH 7. Except for the acid, no other additives are added. Put two aliquots of the protein sample into a glass beaker and put it in a 40°C water bath. Mix with a spatula in one of the beakers to disperse 0.2% (w / w) Neutrase 0.8L (Batch PWNO 1208; Novozymes A / S, Krogshoejvej 36, 2880 Bagsvaerd, Denmark). The samples were incubated at 40°C for 0.5 hours. Both samples were then placed in the refrigerator before measuring viscosity and solubility.

[0041] According to US Patent 6,005,073 (pH 2.8; 10,000 g force), a chicken protein isolate derived from myofibril protein and sarcoplasmic protein was produced from fresh chicken breast muscle and adjusted to pH 5.5 to precipitate the protein. Then sodium hydroxide (2N) was added dropwise to adjust the precipitated protein to pH 8.0. Except for the acid, no other additives are added. Put two aliquots of the protein sample into a glass beaker and put it in a 55°C water bath. Mix with a spatula in one of the beakers to disperse 0.5% (w / w) Alcalase 2.4 L FG, (Batch PLN05212; Novozymes A / S, Krogshoejvej 36, 2880 Bagsvaerd, Denmark). The samples were incubated at 55°C for 1.5 hours. The two samples were then adjusted to pH 7 and placed in the refrigerator before measuring viscosity and solubility.

Abstract

A water soluble peptide composition derived from animal muscle tissue proteins is provided. The peptide composition contains less than about 1 weight percent fats and oils based upon the weight of the peptide composition and less than about 2 weight percent ash based on the weight of the peptide composition.

Description

Background of the invention [0001] The invention relates to a water-soluble protein product and a method for preparing the water-soluble protein product. More specifically, the present invention relates to such a water-soluble product obtained from protein derived from animal muscle tissue as the raw material of the method. [0002] Prior to the present invention, animal proteins have been pre-digested with enzymes to produce peptone products, which can be used as a growth medium for microorganisms. Peptone is a peptide used for bacterial growth. Unfortunately, the composition of these peptone products can vary greatly, depending on the source of animal protein, so that it is difficult for users to achieve reproducible results. Components that can vary greatly include fat and ash (minerals). In addition, these peptone products contain up to about 20% by weight of fats and oils based on the peptone composition, and up to 12% by weight of ash based on the weight of the peptone compo...
