[0029] Example: Collect seawater samples from the surrounding sea area of Xiamen and the inner saline lake—Yundang Lake for pollution detection. The seawater samples collected include Yundang Lake, Baicheng sea area and ferry terminal sea area. The basic parameters of each collected water sample are shown in the table below. The results obtained are attached figure 2 As shown, the exposure of water samples in Yundang Lake and Baicheng to adult male fish caused a significant increase in vitellogenin (relative to clean seawater), indicating that the water samples in these two places were polluted by environmental estrogen.

Abstract

The invention relates to a water estrogen pollution detection method based on oryzias melastigma vitellogenin, and belongs to the field of environmental pollution detection. Currently, no marine fish is used for environment estrogen biological detection, and biological detection technology of seawater estrogen pollution is insufficient. According to the invention, euryhalic male oryzias melastigma with strong adaptability is selected as an in-vivo model; a primer is designed; the expression of vitellogenin (VTG1) is detected by a real-time quantitative PCR method; and 18S ribosome RNA (18S) is used as reference genes to establish a detection system of the VTG1 transcriptional level. Male fish is exposed to 17-beta-estradiol and clean artificial seawater, which are used as a positive and a negative control of the system, and appropriate exposure time is explored. The detection method can be used for detection of environment estrogen pollution degrees of seawater samples, river estuary samples, and saltwater internal lake water samples.

Description

technical field [0001] The invention belongs to the field of environmental pollution detection, and relates to a water body environmental estrogen pollution detection method based on a new type of euryhaline model fish vitellogenin, which can be widely used in the environmental estrogen in areas such as inner lakes in coastal cities, river estuaries, and seawater. Hormone contamination assessment. Background technique [0002] At present, chemical detection methods, such as liquid chromatography and mass spectrometry (HPLC-MS), are mainly used to solve the pollution problem of environmental estrogen. The advantage of these methods is high sensitivity, but the analytical instruments required by these methods are expensive, and the cost is very high, so it is difficult to promote and popularize. In addition, these methods have higher requirements on samples. Environmental samples are different from general samples in that they have the characteristics of a wide variety of co...
