Abstract

The invention provides a separation and purification method for preparing high-purity spinosad, and relates to the technical field of antibiotic extraction. The method steps include: pretreatment of Saccharopolyspora spinosa fermentation broth, leaching, macroporous resin adsorption, nanofiltration concentration and crystallization. The separation and purification process is optimized, and a pretreatment agent, a leaching solvent, an eluent and a crystallization agent are used, and the amount is optimized; precipitated centrifugal precipitate is subjected to secondary leaching to improve the purity and yield of the spinosad, wherein the purity 97.6% to 98.8%, and the recovery rate is 91.3% to 95.8%. In addition, the operation process of macroporous resin adsorption is optimized, the amountof organic solvent is effectively reduced, the production cost is reduced, the production cycle is shortened, and the process flow is simpler and easier to control.
