Embodiment 1

[0027] Example 1: A strain of endophytic fungus isolated from living organisms of Atractylodes macrocephala in Zhejiang Province.

[0028] The endophytic fungus is separated and obtained by the following steps: taking fresh stems and leaves of Atractylodes macrocephala and washing them under clean water to remove surface impurities. Cut stems and leaves of appropriate size, wrap them with gauze, and perform three-step surface disinfection treatment: the volume fraction is 75% ethanol solution for 1 min, the volume fraction is 1% sodium hypochlorite solution for 3 minutes, and the volume fraction is 75% ethanol solution for 30 seconds. Sterilize and take out, cut out small pieces of about 0.5cm×0.5cm in the middle part of stem and leaf tissue, and inoculate them in a sterile separation and culture plate with penicillin (50mg / L), 4 pieces per dish. Seal with parafilm, record the number, and incubate for 7-12 days at room temperature. Regularly observe the growth of the colony, and...

Embodiment 2

[0035] Example 2: Atractylodes macrocephala endophytic fungi were prepared by liquid fermentation of PDB to obtain lactones I and II.

[0036] (1) Strain activation and fermentation: Take AM06 strain preserved at low temperature (4°C), inoculate it in PDA medium, and place it in the dark at 26°C for 5-7 days. When the strains are overgrown with the culture medium, burn the puncher with an alcohol lamp. After it cools, punch 15 bacterial clumps in the denser colonies in the PDA petri dish, and pick them up with the same burned and cooled inoculation needle. The PDB conical flask was sealed and placed in a constant temperature shaker cabinet for fermentation (constant temperature shaker cabinet: 180rpm, 26°C). The above operations are all carried out on the ultra-clean table, and the punch and the inoculation needle need to be burned before and after each use. When the petri dish is punched, the fan-shaped continuous punch is appropriate.

Abstract

The invention discloses Rhizoma Atractylodis Macrocephalae endophytic fungi and application thereof. The endophytic fungi are obtained by separating and purifying endophytic fungi from Rhizoma Atractylodis Macrocephalae living bodies of atractylodes plants of the composite family, and are classified and named as AM06 (Boeremiaexigua), and the strain deposit number is CGMCC No. 17782. According to the invention, aractylenolide I and aractylenolide II are produced by liquid fermentation of an ophytic fungus strain PDB. The endophytic fungi of Rhizoma Atractylodis Macrocephalae is an important microorganism for searching new resources of the aractylenolide I and the aractylenolide II, and has high application value.

Description

Technical field [0001] The invention relates to the technical field of microorganisms, in particular to an endophytic fungus of Atractylodes macrocephala and its application in the preparation of Atractylodes lactone I and II. Background technique [0002] Atractylodes Ⅰ and Atractylodes Ⅱ are the main active ingredients of Atractylodes macrocephala, and they are tricyclic sesquiterpene lactone compounds contained in Atractylodes macrocephala. Modern pharmacological research shows that Atractylodes lactone I and Atractylodes lactone II have various pharmacological activities such as anti-inflammatory, anti-tumor, improving spleen and stomach, inhibiting muscle contraction, preventing Alzheimer's disease, and improving memory. Since the separation of Atractylodes Ⅰ and Atractylodes Ⅱ from the volatile oil of Atractylodes in the 1980s, people have tried to optimize various extraction conditions and increase their yield. Some scholars and others extract the volatile oil from Atract...
