Embodiment 1
[0025] Embodiment one: Type VI collagen purification:
[0026] Amniotic membranes were isolated from fresh human placenta and washed several times with 0.5M sodium acetate to remove serum proteins. The washed amnion was minced and homogenized. Each 80 g (wet weight) of amniotic tissue was treated with 8 mg of pepsin and 0.5 M formic acid (pH 2.1) for 24 hours, and the whole process was carried out at 4 degrees Celsius. After centrifugation, the supernatant was collected, and a NaCl solution having a final concentration of 1.2 M was added to the solution to collect the collagen component as a precipitate. The pellet was redissolved in 0.05m Tris-HCl buffer (pH 8.0, containing 1M NaCl), and dialyzed against 0.5M acetic acid solution containing 1.2M NaCl. The precipitate was removed and the supernatant was further dialyzed against 1.7M NaCl in 0.5M acetic acid. The resulting pellet was resuspended in a solution containing 0.2% SDS, 1 mM EDTA, 50 mM Tris-HCl, 100 mM NaCl, pH 7....
Embodiment 2
[0029] It is basically the same as Example 1, except that type VI collagen complexes are isolated from fresh rat placenta.
Embodiment 3
[0031] It is basically the same as in Example 1, except that type VI collagen complexes are isolated from fresh mouse placenta.
