The optimal extraction process comprises the following steps: single-factor experiment, response surface design, determination of optimal extraction conditions of polyphenol in the tinder fungus, and determination of crude extract polyphenol content,. wherein the single-factor experiment comprises drawing of a standard curve, influence of ethanol concentration on extraction of tinder fungus polyphenol, influence of material-liquid ratio on extraction of the polyphenol in the tinder fungus, influence of ultrasonic time on extraction of the polyphenol in the tinder fungus, influence of ultrasonic temperature on extraction of the polyphenol in the tinder fungus, influence of extraction times on extraction ofthe polyphenol in the tinder fungus, and multiple experimental principle steps. According to the optimal extraction process of the polyphenol in the tinder fungus, the influence of the ethanol concentration, the material-liquid ratio, the ultrasonic time, the ultrasonic temperature and the extraction frequency on extraction of the polyphenol in the tinder fungus is determined by designing a single-factor experiment, and the optimal extraction process is obtained by designing a response surface. Reference is provided for development and utilization of the polyphenol in the tinder fungus.
