[0045] Example 1: Treatment of SJL RCS Tumors with LT Pathway Inhibitors Reduces Total LN / Tumor Volume

[0046] SJL mice were treated with 0.3-0.4 mg of mouse LTBR-hIgG1 fusion protein via intraperitoneal administration, 3 days before tumor transplantation (D-3), on the day of transplantation (D0), or at the time of transplantation. Performed 3 days later (D3). Tumor transplantation was basically performed according to the literature 9 . 5×10 6 RCS cells depleted of T cells were injected intravenously to inoculate and grow in the organ. After 5-7 days, mesenteric, brachial and axillary LNs were dissected and their weights calculated as a percentage of total body weight. Table I shows that the LN size decreased in all experiments. Spleen size was also reduced in 1 of 3 trials, but the reduction in spleen weight was not significant. The reduction in LN weight ranged from approximately 50% with single dose treatment to 80-90% with multiple dose treatment.

Application of lymphotoxin pathway inhibitors in tumor therapy, especially follicular lymphoma.

Description

[0001] The present invention relates to compositions and therapeutic uses of lymphotoxin pathway inhibitors for the treatment of tumors, especially germinal center-derived lymphomas (follicular lymphomas). Background of the invention [0002] Tumor necrosis factor (TNF)-related cytokines can both mediate host defense and immune regulation. Members of this family exist as membrane anchors, acting locally through direct cell-to-cell contact, or as secreted proteins that can diffuse to more distant targets. The corresponding family of receptors signal the presence of these molecules, resulting in the initiation of cell death or proliferation and differentiation in the target tissue. So far, at least 11 receptor-ligand pairs have been recognized in TNF ligand and receptor families, including: TNF: TNF-R; LT-α: TNF-R; LT-α / β: LT-β- R; FasL: Fas; CD40L: CD40; CD30L: CD30: CD27L: CD27; OX40L: OX40 and 4-1BBL: 4-1BB.
