Embodiment 1

[0017] Example 1: Utilizing microbial flora to treat petroleum-containing wastewater

[0018] (1) Bacillus and Enterobacter were selected for slant culture, first-level and second-level seed culture in LB medium respectively, with a culture temperature of 29°C and a culture time of 20 to 36 hours;

[0019] (2) Collect the cells obtained in step (1) respectively, and wash them twice with normal saline; then reconstitute the OD with normal saline 620nm To achieve a bacterial suspension between 11 and 12, mix the Bacillus and Enterobacter suspension at a volume ratio of 2:1 to construct an artificial flora;

[0020] (3) Put the artificial flora constructed in step (2) into the wastewater containing 100 grams of oil per liter according to the inoculation amount of 6% to 8% by volume, stir at 33°C for 10 days, and detect the degradation rate of crude oil To judge the treatment results, the oil-containing wastewater was treated synchronously without adding bacterial suspension as a...

Abstract

The invention discloses a treatment method of wastewater containing petroleum by utilizing a microorganism flora. The method comprises the following steps: culturing bacillus and enterobacter to respectively obtain strains; cleaning, and re-preparing bacterium suspension with OD620nm between 10 and 12 with normal saline; mixing the bacillus and enterobacter to construct a manual flora in a volume size of (2-3):1; and inoculating the constructed manual flora into petroleum wastewater in a inoculation amount of 6 to 8 percent of the volume ratio to realize treatment to the petroleum containing wastewater. Experiments prove that the crude oil degradation rate of treating petroleum containing wastewater by utilizing microorganism flora is remarkably higher than that of a single strain, and foundation is established to large-scale application of artificially constructed flora in wastewater containing petroleum treatment.

Description

technical field [0001] The invention relates to a method for treating petroleum-containing wastewater, in particular to a method for utilizing microbial flora to treat petroleum-containing wastewater. Background technique [0002] Due to various reasons, soil and water resources are polluted to varying degrees during the process of mining, transporting, refining and using petroleum. According to investigations, the area of contaminated soil in the oil development zone of Daqing Oilfield exceeded 75%, and the area of contaminated soil in the agricultural development zone exceeded 20%. At present, the average concentration of oil in my country's offshore waters has reached 0.055mg / L. Leaked oil and wastes such as sludge and oily wastewater generated during the refining process enter the soil and cause damage to the structure and function of the soil ecosystem, thereby poisoning plant growth, destroying surface vegetation, and reducing food production; at the same time, se...
