[0033] Example 1, the discovery of pAcTRT1 and its distribution characteristics on the chromosome of wheatgrass

[0034] 1. Discovery of pAcTRT1

[0035] The 6P short arm DNA sequence fragment was obtained by microdissection and DOP-PCR using wheat-Agropyron 6P short arm system as material. These sequences were connected to the pMD19-T vector and introduced into Escherichia coli to obtain a DNA sequence fragment library of the 6P short arm. Differential DNA clones between wheatgrass Z559 and wheat Fukuho were screened by dot hybridization and sequenced.

[0036] According to BLAST analysis, there is a DNA fragment with a length of 962bp and a GC content of 44.3%. The DNA fragment is named pAcTRT1, as shown in sequence 1 of the sequence listing.

[0037]pAcTRT1 shares 90% sequence similarity with the tandem repeat sequence pSc200 in rye.

[0038] 2. The distribution characteristics of pAcTRT1 on the chromosome of Agropyron chinensis

[0039] Using pAcTRT1 as a probe, two di...

Abstract

The present invention discloses an agropyron cristatum chromosome end portion specific tandem repeat sequence and application thereof, and the agropyron cristatum chromosome end portion specific tandem repeat sequence (being a molecular marker, named as a specific DNA molecule) is a DNA molecule as shown in sequence 1 of a sequence table. The present invention also provides the application of the specific DNA molecule in identifying whether a genome of a plant material to be tested contains genetic materials derived from agropyron cristatum chromosome end portion. Significances are as follows: (1) by obtaining pAcTRT1, agropyron cristatum chromosome identification is easier and more accurate, and a blank of no ideal probe available for agropyron cristatum chromosome identification can be filled; and (2) the pAcTRT1 can be used for detecting P chromatin of the agropyron cristatum chromosome end portion under wheat background.

Description

technical field [0001] The invention belongs to the field of biotechnology, and in particular relates to a specific tandem repeat sequence at the end of wheatgrass chromosome and its application. Background technique [0002] DNA repeats are a major component of most plant genomes. It can be divided into two categories: tandem repeats and scattered repeats. Tandem repeats are the main components of higher plant genomes and are distributed in specific regions of chromosomes, such as centromeres, telomeres, heterochromatin regions, etc. Some tandem genes are considered to provide a large number of protein or RNA products with important physiological and biological functions. Changes in tandem repeats can be used to study the evolution of plant genomes and determine evolutionary trends. [0003] Chromosome identification is an important link in the analysis of genome relationships. Fluorescence in situ hybridization (FISH) technology has a unique advantage in identifying ch...
