Embodiment 1

[0016] A method for organically solidifying organic tissue culture Aureus aureus, comprising the steps of:

[0017] (1) Remove the stems of the roots and leaves of wild clematis as explants, wash the surface, sterilize in 75% ethanol for 30 seconds, and then sterilize with 0.1wt.% mercuric chloride for 7 minutes. Rinse with bacterial water 4 times;

[0018] (2) Under sterile conditions, cut the stem into lcm-long segments with 1 axillary bud, inoculate them on the induction medium, cut the subcultured stems into lcm, move them into the organic solidified matrix, and continuously light them for 12 hours every day. The light intensity is 1500lx, the culture temperature is 25±1℃, and cultured for 120 days to form effective roots, promote the thickening of bud diameter and the differentiation of stems and leaves; the induction medium is MS basic medium supplemented with 6-BA1.0mg / L, NAA0 .1mg / L and ZT0.02mg / L.

[0019] The organic solidification matrix contains 5g / L white sugar,...

Abstract

The invention relates to a method for tissue culture of anoectochilus formosanus through an organic solidified medium. Wild anoectochilus formosanus is used as an explant, a stem is obtained after roots and leaves are removed, after the stem is subjected to surface cleaning, the stem is placed in 75% ethyl alcohol to be sterilized for 30 seconds, 0.1 wt.% of mercury bichloride is used for sterilization for 7-10 min, and the stem is washed four or five times with sterile water; under the sterile condition, the stem is cut into a segment which is 1-2 cm in length and has one or two axillary buds, an inducing medium is inoculated with the segment, continuous illumination is conducted for 12 h every day, illumination intensity is 1,500-3,000 lx, the culture temperature is 25+ / -1 DEG C, culture is conducted for 120-125 days, effective roots are formed, and increase of the bud diameter and differentiation of a stem and leaves are promoted. A high-quality organic seedling is cultured and has a complete root system, the stem and the leaves, and the survival rate is high.

Description

technical field [0001] The invention belongs to biological cell tissue technology, and in particular relates to a method for tissue-culturing Aurantia clematis with an organic solidification mechanism. Background technique [0002] Clematis clematis, also known as golden clematis and golden silk grass, is a rare perennial Chinese herbal medicine of the Orchidaceae genus Molophyllum genus. In 1990, it was listed as an endangered medicinal plant by the Fujian Provincial Government, which mainly produces this medicinal material. It is widely used among the people, and has the effects of clearing away heat and cooling blood, dispelling wind and dampness, detoxifying, relieving pain, and antitussive. It has remarkable effects in treating children with convulsions, removing fetal poison, protecting the liver, treating diabetes, breast cancer, and gastric cancer. Known as "King of Medicine", "Golden Grass", "Miraculous Medicine", "Bird Ginseng" and other laudatory titles. Accordi...
