Embodiment 1

[0081] This effect was confirmed in the ovalbumin allergy mouse model experiment of Example 1. In this example, groups of weaned mouse pups were fed (i) water, (ii) - HA formula or (iii) TGF-beta enriched -HA formula. Scheme (iii) is a composition according to an embodiment of the present invention. - HA contains all 5 peptides SEQ ID1-5. Mice pups in each of the three regimen groups were sorted according to whether they were suckled by a naive (not exposed to ovalbumin) dam or by an immunized dam that had been exposed to ovalbumin (OVA) further divided into two subgroups.

[0082] Mouse pups were then subjected to a food allergy induction protocol, briefly consisting of intraperitoneal injection of ovalbumin / aluminum hydroxide (adjuvant), followed by oral OVA challenge. They were then measured for symptoms of allergy, especially diarrhea (Experiment 1). A second set of OVA challenge (Experiment 2) was performed and allergy symptoms, especially diarrhea, were again mea...

Abstract

This invention relates to the administration of a combination of TGF-Belta and one or more peptides comprising specific sequences of Belta-lactoglobulin. The peptides are hypoallergenic but retain thecapability to induce tolerance to the native protein. The combination of TGF-Belta and one or more of these peptides has the therapeutic effect of inducing and maintaining oral tolerance towards newly introduced proteins, in particular, at the weaning period. The combination can modulate the infant and toddler immune response to allergens.

Description

technical field [0001] The present invention involves the administration of TGF-[beta] in combination with one or more peptides comprising specific sequences for [beta]-lactoglobulin. The peptides are hypoallergenic but retain the ability to induce tolerance to native proteins. The combination of TGF-β with one or more of these peptides has a therapeutic effect in inducing and maintaining oral tolerance to newly introduced proteins, especially during the weaning period. The combination modulates the immune response of infants and toddlers to allergens. Background technique [0002] Food allergies (often the first in life to cow's milk) are in most cases caused by a reaction to proteins in the food. During the first few years of life, the immune system is still developing and may not be able to develop tolerance to dietary antigens (this can also be described as insufficient induction of oral tolerance). As a result, the infant or child or juvenile animal develops an exagg...
