Problems solved by technology

In the introduction and breeding, it was found that the adults of rock oyster and long oyster are obviously different, but the spat are not easy to distinguish
Due to the strong plasticity of the shell of oysters and the overlapping of distribution areas, there are strong subjective factors in the identification of oyster species only based on external morphological characteristics, which brings great difficulties to the development of new species of rock oyster.

Embodiment Construction

[0020] The present invention is further explained and illustrated by specific examples below.

[0021] The rock oysters and long oysters used in this example were taken from Shandong Rongcheng Breeding Base. Eighteen individuals of each type of oyster were selected, for a total of 36 individuals.

[0022] Log in to the National Center for Biotechnology Information (NCBI, http: / / www.ncbi.nlm.nih.gov / ), and download the existing mitochondrial COI sequences of Oyster oyster and Oyster oyster (Oyster oyster Genbank accession number: HM015198, oyster oyster Genbank accession number is: AB033687).

[0023] The downloaded sequences were compared using Mega6.0 software.

[0024] The primer design software Primer Premier 5 was used to design primers in relatively conserved regions of the sequence. The designed COI primer sequence is:

[0025] COIY-F: 5'-CTATTAGAAATATGCGGTCTG-3',

[0026] COIY-R: 5'-AACAAAGTAAGTATCGTGAAG-3'.

[0027] The DNA of rock oyster and oyster oyster was ex...

Abstract

The invention relates to the field of molecular biology technology, and discloses a PCR-RFLP identification method for rock oyster and oyster oyster and a pair of COI primers for identification. The 16S rRNA and COI gene fragments of two oysters were amplified by using the 16S rRNA general primer and the COI primer respectively, and the amplified target gene fragments were sequenced, and the restriction endonuclease specific site analysis was performed on the sequencing results , finally select the same restriction endonuclease Alu Ⅰ Digest the PCR product, and use 2.0% agarose gel to detect the digestion result of the PCR product. The results showed that the PCR products of rock oyster and long oyster Alu Ⅰ Digestion produced fragments of different sizes, and the rock oyster and long oyster could be identified according to the restriction map. This simple, reliable and economical identification method can provide technical means for the germplasm identification of rock oyster and long oyster.

Description

technical field [0001] The invention belongs to the technical field of molecular biology, and in particular relates to a PCR-RFLP identification method for mitochondria of rock oysters and long oysters and a pair of mitochondrial COI gene primers for identification. Background technique [0002] Oyster (Ostreidae) is a species widely distributed in the world. It is an important aquaculture object in the mariculture industry of various countries in the world, and it is also the economic shellfish with the largest output in my country and even in the world. In 2016, China's oyster farming output reached 4.83 million tons, ranking first in the world. Oysters of the genus Colossus are important economic species in my country, and the common species are mainly long oysters ( Crassostrea gigas ), Omi oysters ( Crassostrea ariakensis ), Hong Kong Giant Oyster ( Crassostrea hongkongensis ), Portuguese oysters ( Crassostrea angulata ), and Kumamoto oysters ( Crassostrea sika...
