Embodiment Construction

[0030] Below in conjunction with accompanying drawing and experimental data, the present invention is further explained and illustrated

[0031] 1. Materials and methods

[0032] Cell lines and drug treatment, qRT-PCR analysis, Western blot analysis, immunohistochemical assay, MTT assay, colony formation assay, apoptosis analysis cells, cell cycle analysis, the above methods are all existing methods, and will not be repeated here .

[0033] 2. Patient tissue samples

[0034] In this study, patients from the Third Xiangya Hospital of Central South University and the Cancer Hospital Affiliated to Xiangya School of Medicine, Central South University were used. Paraffin-embedded specimens of 112 patients with histopathologically diagnosed bladder cancer (BUC) who underwent radical cystectomy (RC) in 2009 (Table 1). Thirty radical cystectomy (RC) BUC tissue specimens and corresponding adjacent non-neoplastic tissue specimens were immediately stored at −80°C and used for qRT-PC...

Abstract

The invention discloses application of SPAG5 as a target site in preparing urinary bladder carcinoma treating medicine. Researches show that SPAG5 up-regulation can be frequently detected in primary BUC tissues, and increasing and reducing SPAG5 expression can respectively enhance or inhibit in-vitro and in-vivo BUC cell proliferation and can respectively inhibit or promote in-vitro and in-vivo cell apoptosis. Therefore, the SPAG5 can serve as the target site to be applied to preparing the urinary bladder carcinoma treating medicine and further can be applied to preparing urinary bladder carcinoma molecular markers and preparing urinary bladder carcinoma relapse predicting molecular markers. Mechanism researches show that the SPAG5 can promote BUC proliferation and inhibit BUC apoptosis mainly by activating AKT / mTOR signal channels and at least partially increasing Wnt3. Therefore, the SPAG5 can be applied to preparing target negative regulating Wnt3 expression preparation. Meanwhile, the Wnt3 also can serve as a target site to be applied to preparing urinary bladder carcinoma treating medicine.

Description

technical field [0001] The invention belongs to the technical field of biomedicine, and specifically relates to the application of SPAG5 as a target site in the preparation of drugs for treating bladder cancer. Background technique [0002] Bladder urothelial carcinoma (BUC) is the fourth most common tumor in men and the seventh most common in women, affecting more than 400,000 people worldwide each year. Radical cystectomy (RC), with or without preoperative RT, offers the best chance for cure in muscle-invasive bladder urothelial tumors and has also been instrumental in the treatment of non-muscle-invasive bladder urothelial tumors . Despite recent developments in treatment strategies including radical cystectomy and radiotherapy, bladder urothelial tumor recurrence occurs in approximately 50% of patients due to heterogeneity in disease characteristics. Given this situation, an accurate prognostic and predictive assessment after radical cystectomy is required. This is im...
