[0024] 1 Extraction of mycelium genomic DNA:

[0025] Genomic DNA was extracted according to the instructions of the high-efficiency plant genomic DNA extraction kit. DNA purity was checked using a spectrophotometer.

[0026] 2 Design of LAMP-specific primers

[0027] The translation elongation factor alpha 1 gene (TEF1) gene, which is highly conserved in gene and expression regulation in different species, was selected as the target gene. Find and download the TEF1 gene sequence of G. anthracnose and its related species from the NCBI website, use BioEdit software for sequence comparison, analyze and find the specific conserved sites of G. anthracis, and then use PrimerExplorer V5 (http: / / primerexplorer.jp / lampv5e / index.html) online software to design primers (Table 1).

Abstract

The present invention provides a LAMP detection primer for detecting C. gloeosporioides in olive tissue. The primer has a sequence as shown in SEQ ID NO. 1-4. Based on an LAMP technology, a new targetgene is selected for designing and screening primers, so that the technology can be more accurately used for detecting C. gloeosporioides, and provides technical support for timely prevention and treatment of olive anthracnose.

Description

technical field [0001] The invention belongs to the field of phytopathology, and in particular relates to a method for rapidly detecting glyospora anthracnose in olive tissues. Background technique [0002] Glosospora anthracnose ( C. gloeosporioides ) has a wide variety of hosts, including more than 2,200 species of plants, which cause anthracnose and cause serious economic losses to agricultural production. The classification and identification of traditional pathogenic bacteria are mainly based on morphological characteristics and pathogenicity determination, etc. However, the morphological variation between anthrax species and even strains within a species is large, and the taxonomic characteristics of similar species are slightly different, making the classification of this genus difficult to operate . As a main and important species in this genus, G. glyospora anthracnose has the characteristics of aggregated populations and compound species in terms of morphological...
