Based on the formation process of the initial water saturation of the compact sandstone reservoir, the initial water saturation of the compact rock core is established by adopting a water increasing method, a valve is additionally arranged on a plunger at the outlet end of a rock core holder through pipeline volume calibration, and the valve is closed when the initial water saturation is established, and formation water or simulated formation water required for establishing the initial water saturation of the compact rock core can completely enter the rock core. By monitoring the resistivity of the rock core, formation water or simulated formation water injected into the rock core is ensured to be uniformly distributed in the rock core, and vacuumizing, porosity measurement and initial water saturation establishment are creatively combined.
