Abstract

The invention provides a method and a system for detecting concentration of predetermined ions in cells, the method comprises the following steps: irradiating to-be-detected cells through a light beam to make predetermined fluorescent proteins in the to-be-detected cells generate a feedback light beam under irradiation, and the predetermined fluorescent proteins having sensitivity of the predetermined ions; collecting light intensity information of the feedback light beam at the preset wavelength; and determining the concentration of the predetermined ions in the to-be-detected cells based on the light intensity information and a predetermined calibration curve of the predetermined ions. According to the invention, the non-invasive detection of the absolute concentration of the predetermined ions in the cells is achieved by constructing the specific ion-sensitive fluorescent protein sensor, and the ion concentration of the predetermined ions in the cells can be accurately detected according to the comparison between the light intensity information of the feedback light beam of the fluorescent proteins under the light beam and the predetermined calibration curve; the accuracy of concentration detection is improved, and meanwhile, the method has the characteristics of real-time detection and dynamic response.

Description

technical field [0001] The present disclosure relates to the technical field of biotechnology, in particular to a method and system for detecting the concentration of predetermined ions in cells. Background technique [0002] The ion concentration and pH of various element ions in the cell play a vital role in the physiological form, function, and metabolism of the cell itself and its subcellular structure. Taking the concentration of chloride ion in halogen elements as an example, it plays an important role in maintaining the cell membrane potential, pH gradients, as well as the function of numerous organelles (such as mitochondria) can all be affected. Existing technologies that rely on sensitive films, electrodes, and chemical dyes to detect intracellular ion concentrations cannot meet the requirements of real-time and dynamic detection due to problems such as sensor physical size, manufacturing cost, cytotoxicity, and single-cell measurement accuracy. Moreover, although...
