The preparation method comprises the following steps of: decocting and extracting the scutellaria baicalensis, adjusting the pH value of the decocted mixture by using sodium hydroxide solution in each decocting process, mixing the obtained decoction liquid so as to obtain liquid A; adding water into the bupleurum for distillation, collecting distillate produced in the first 30 to 40 minutes so as to obtain liquid B; adding water into the produced dregs of a decoction of the bupleurum and the lonicera confusa for decoction, mixing the obtained two-time decoction liquid and the residual liquid of the bupleurum subjected to distillation, concentrating and filtering the obtained liquid, and adjusting the PH value of the obtained filter liquid so as to obtain liquid C; and finally, mixing 30 to 35 volume percent of liquid A, 5 to 10 volume percent of liquid B and 55 to 60 volume percent of liquid C, and then carrying out separated-loading and sterilization on the obtained liquid so as to obtain the micro-enema.
