Embodiment 1

[0043] Embodiment 1, the method for cultivating the transgenic wheat that iron content improves in grain

[0044] 1. Experimental materials

[0045] 1. Construction of pBAC47P vector:

[0046] The pSP72 vector of Promega Company was used as the starting vector, the promoter of wheat high molecular weight glutenin subunit 1Dx5 was inserted between HindIII and Sal I, and the NOS terminator was inserted between Kpn I and EcoR I. The NOS terminator comes from the pBI121 vector of Clontech Company in the United States. The 1Dx5 promoter is derived from the plant expression vector pBPC30.

[0047] The nucleotide sequence of the promoter of wheat high-molecular-weight glutenin subunit 1Dx5 is shown as nucleotides 1-857 at the 5' end of SEQ ID NO:3. The nucleotide sequence of the NOS terminator is shown as nucleotides 1625-1887 at the 5' end of SEQ ID NO:3.

Abstract

The invention discloses a method for cultivating transgenic wheat with seeds with improved iron content. The method for cultivating a transgenic plant with improved iron content comprises a step of importing an expression cassette consisting of an endosperm-specific promoter, an encoding gene of soybean ferritin and a terminator into a starting plant to obtain a target transgenic plant with the iron content higher than that of the starting plant. The experiment proves that the iron content in the seeds of the transgenic wheat is remarkably improved and the transgenic wheat with high iron content is obtained. The method has significance for improving the nutritional quality of wheat and the daily diet structure of people.

Description

technical field [0001] The invention relates to a method for cultivating transgenic wheat with increased iron content in grains. Background technique [0002] Iron is an indispensable and important element in many normal physiological processes of the human body. The lack of human iron nutrition has become one of the most serious nutritional deficiencies in the world today. According to a report provided by the Standing Committee on Nutrition of the United Nations System in 2004, more than 1 / 2 of the world's population is affected by malnutrition (WHO, 1992). It is estimated that 2 billion people in the world suffer from iron deficiency, most of whom are women and children (Long Xinxian, 1998). Our government is also very concerned about the mineral deficiency of vulnerable groups. [0003] Adults contain about 3-5g of iron, which is the most abundant trace element in the human body. 70% of iron exists in hemoglobin and myoglobin, 25% of iron exists in the form of ferrit...
