Embodiment

[0198] Strain Construction: All strains used in this study are listed in Table 1. Use JCL16 (rrnB T14 ΔlacZ WJ16 hsdR514ΔaraBAD AH33 ΔrhaBAD LD78 / F'[traD36proAB+lacI q ZΔM15]) as wild type (WT) (Atsumi et al., 2008). All plasmids were propagated using XL-1Blue (Stratagene). Enzymes were expressed using BL-21DE3 (Invitrogen) prior to enzyme assays. Gene deletion was performed by P1 transduction using single gene knockout strains from the Keio collection (Baba et al., 2006). Each gene knockout was verified by PCR using the following primers flanking the deletion locus: gltA(5'-

Abstract

Provided are microorganisms and plants that express or overexpress enzymes that catalyze the conversion of a four carbon metabolite (malate) to acetyl-CoA. Also provided are methods of generating such organisms and plants and methods of synthesizing biomass, biofuel, oil, chemicals and biochemicals using such organisms and plants.

Description

[0001] Statement Regarding Federally Funded Research [0002] This invention was made with government support under Grant Nos. DE-AR0000085 and DE-AR0000201 awarded by the U.S. Department of Energy. The government has certain rights in this invention. [0003] Cross References to Related Applications [0004] This application claims priority to US Provisional Application Serial No. 61 / 841,310, filed June 29, 2013, the disclosure of which is incorporated herein by reference in its entirety. technical field [0005] Metabolically modified microorganisms and plants and methods of producing such organisms and plants are provided. Also provided are methods of producing chemicals by contacting a suitable substrate with a metabolically modified microorganism or plant of the disclosure and an enzyme preparation. Background technique [0006] Acetyl-CoA is a central metabolic key to both cell growth and the biosynthesis of a variety of cellular components and products, including f...
