Problems solved by technology

[0004] At present, there is a lack of research on the genetic variation of the microRNA-320a-1 gene in cattle. The functional research on the polymorphic site of the gene and its genetic variation and growth and development traits (such as: weight, body height, body length, chest width, chest depth, and chest circumference, etc. traits) association research is still blank

Abstract

The invention discloses a detection method of single nucleotide polymorphism of a Qinchuan cattle microRNA-320a-1 gene and an application thereof. The detection method comprises the following steps: with the to-be-detected Qinchuan cattle whole-genome DNA as a template and a primer pair P2 as primers, performing PCR amplification of a Qinchuan cattle microRNA-320a-1 gene segment; digesting the PCR amplification product with restriction endonuclease PuvII, and performing agarose gel electrophoresis of the amplified segment after the enzyme digestion; and identifying the single nucleotide polymorphism of Qinchuan cattle microRNA-320a-1 gene rs518926539:C>T according to the result of agarose gel electrophoresis. The detection method disclosed by the invention lays a foundation for the marker assisted selection of the growth character for Qinchuan cattle meat based on the relationship between the microRNA-320a-1 gene SNP and the growth character, and is favorable for quickly establishing a Qinchuan cattle population with good genetic resources.

Description

technical field [0001] The invention belongs to the field of biotechnology and relates to the detection of gene single nucleotide polymorphism (SNP), in particular to a method for detecting the single nucleotide polymorphism of Qinchuan cattle microRNA-320a-1 gene. Background technique [0002] Single nucleotide polymorphism (Single Nucleotide Polymorphism, SNP) is the variation caused by the substitution of a single nucleotide (A / T / C / G) in the DNA sequence of the biological genome. Studies have shown that single nucleotide substitutions in the genomic DNA sequence will cause changes in the DNA sequence or amino acids in encoded proteins, which will cause organisms of different genotypes to exhibit different traits. [0003] MicroRNAs are a class of non-coding RNAs about 22 nt in length produced by the transcription of biological genome DNA. Studies have shown that the way microRNAs function is to pair with the mRNA bases of downstream target genes, causing the degradation o...
