Embodiment 1

[0045] In this experiment, 334 1.5-year-old barby barbs were used to stock and raise at the same time in the same pond. The specific steps are as follows:

[0046] (1) Cut off a little tail fin of each barb and then put a label on the barb. And label the collected caudal fins, one-to-one correspondence with each barb, and mark them with Arabic numerals.

[0047] (2) Take 2 mg of caudal fin from each barb barb, then add 50 μl of universal one-step DNA extraction solution (Sangon Bioengineering (Shanghai) Co., Ltd.), and incubate at 80° C. for 15 minutes. After brief shaking and mixing, the lysate was taken directly for PCR.

[0048] The PCR system is as follows:

[0049]

[0050] The reaction conditions of PCR were as follows: pre-denaturation at 94°C for 2 minutes; 30 cycles of denaturation at 94°C for 20 s, annealing at 51°C for 20 s, and extension at 72°C for 25 s; and extension at 72°C for 5 min.

[0051] (3) Pipette 4 μl of the obtained PCR product and use 1% agarose...

Abstract

The invention discloses a method for rapidly screening rapid growth individuals of spinibarbus hollandi, a kit and an application. The 686th nucleotide and the 522th nucleotide of a promoter of a melanocortin receptor of the spinibarbus hollandi are named as SNP1 and SNP2 separately and are detected; an SNP1 site is a rapid growth group when being a CT heterozygote, is a subrapid growth group when being a C homozygote, and is a disadvantaged group when being a T homozygote; and an SNP2 bit is a rapid growth group when being an AG heterozygote, is a subrapid growth group when being a G homozygote and is a disadvantaged group when being A homozygote. Due to an additive effect of a minor gene, the growth speed is the highest when both the SNP1 site and the SNP2 site are heterozygotes; the growth speed is the second when only one of the two sites is the heterozygote and the other one is the homozygote with a relatively high growth speed, and so on. According to the method, the kit is obtained, and the method is used for rapidly screening the rapid growth individuals of the spinibarbus hollandi.

Description

technical field [0001] The invention belongs to the technical field of aquatic genetics and molecular breeding, and in particular relates to a method, a kit and an application for rapidly screening fast-growing individuals of the barbed barb. Background technique [0002] The barbed barb (Spinibarbus hollandi) belongs to Cyprinidae, Cyprinidae, Spinibarbus subfamily, and the genus Spinibarbus hollandi. It has the advantages of large individual, fast growth, wide appetite, low oxygen resistance, strong disease resistance, and easy breeding. Distributed in the Yangtze River, Qiantang River, Minjiang River, Jiulong River, Pearl River, Yuanjiang, Taiwan Island and Hainan Island and other water systems. Due to the inbreeding and indiscriminate hybridization of the photobarbed barb, the germplasm resources of the photobarbed barb are destroyed, causing the species to degrade, resulting in reduced production, susceptibility to diseases, etc. The germplasm improvement of the photob...
