Problems solved by technology

Using RNAi technology to silence the Me-pel-1 of the 2nd instar larvae of M.enterolobii, it was found that after the Me-pel-1 gene was disrupted, the infection rate of the 2nd instar larvae of M.enterolobii to tomato was significantly reduced

Embodiment Construction

[0047] In order to make the object, technical solution and advantages of the present invention more clear, the present invention will be further described in detail below in conjunction with the examples. It should be understood that the specific embodiments described here are only used to explain the present invention, not to limit the present invention.

[0048] M.enterolobii has been reported successively in recent years, and it has spread all over Hainan Island. It mainly harms cucurbitaceous vegetables, solanaceous vegetables, southern medicine plants and tropical fruit trees, etc. It is gradually replacing root-knot nematode in southern China and has become the most important species in tropical and subtropical regions of China. species of plant root-knot nematodes. Studying the loss of function of growth and development-related genes such as feeding, fitness and survival, which prevents it from infecting host plants, plays an important role in the prevention and control...

Abstract

The invention belongs to the field of bioinformatic analysis technology and discloses an enterolobium cyclocarpum root transcriptome database, fusion protein, a soaking system and a silencing system. The database is composed of more than 38,000 Unigenes sequences, and bioinformatic analysis is utilized to predict potential RNAi target candidate genes of more than 230 enterolobium cyclocarpum root-knot nematodes. A prokaryotic expression technology is utilized to obtain ME-MAPK1, ME-COL1 and MeHsp70 recombinant protein, and a foundation is laid for protein characteristic analysis; the M.enterolobii in-vitro RNAi soaking system and the VIGSRNAi in-vivo silencing system with a tobacco rattle virus (TRV) carrier being a medium are successfully constructed, and Me-mapk1, Me-crt1 and Me-cbp1 gene down-regulated expression is induced; and a necessary molecular foundation is laid for M.enterolobii prevention and treatment.

Description

technical field [0001] The invention belongs to the technical field of bioinformatics analysis, and in particular relates to a transcriptome database, a fusion protein, a soaking system and a silencing system. Background technique [0002] Root-knot nematode Meloidogyne enterolobii was first discovered and named by Yang Baojun et al. on the Meloidogyne enterolobii in Danzhou, Hainan in 1983. Xu Jianhua and others proved that M. mayaguensis, which is widely reported by foreign scholars, is a synonym of M. enterolobii. The nematode is distributed in the Americas, Africa, Europe, etc., and is recognized as one of the most harmful root-knot nematodes in the world. In 2008, M. enterolobii was reported for the first time in Europe. In the same year, EPPO reported that the nematode was intercepted on roses exported from China to Europe. EPPO conducted a risk assessment of the nematode and believed that it was a greater threat to crops in Central Europe. At present, the countries ...
