The method comprises the following steps: preparing a diluent; processing fresh seminal fluid; cooling, balancing, freezing, and unfreezing. A composition, which has a certain concentration and is composed of graphene oxide and glycerin, is added into a freezing liquid to prevent harms caused by formation of ice crystals on sperms; before the seminal fluid is frozen, the seminal fluid is cooled for 20-40 minutes by an ultra low temperature refrigerator with a temperature of -80 DEG C in advance; the difficultythat in a conventional method, seminal fluid should be cooled by a pricy programmed freezing instrument is solved; after freezing, the frozen seminal fluid is unfrozen by a water bath with a temperature of 60 DEG C; the living rate of sperms is effectively improved after the freeze preservation of donkey seminal fluid, and the method is suitable for artificial insemination of donkeys in grass-rootorganizations.
