An enrichment device comprises arefrigeration shell and enrichment collecting devices coaxially arranged in the refrigeration shell; the refrigeration shell comprises two oppositely-arranged half shells, a water pipe penetrating hole is formed in the middle of the refrigeration shell, and a cooling device is arranged between the water pipe penetrating hole and the inner wall of the shell; and the enrichment collecting devices are provided with annular clamping buckles, and filter membranes and sand core filters are clamped in the middles of the annular clamping buckles. The underwater microorganism sample collecting devicecomprises a water inlet pipe, a water outlet pipe and one or more enrichment pipelines connected between the water inlet pipe and the water outlet pipe in parallel; and the enrichment pipelines are composed of first valves, the enrichment collecting devices and flowmeters which are connected in series through pipelines. The underwater microorganism sample collecting method comprises the followingsteps that underwater microorganisms are collected through the filter membranes in real time, and the flow and the flow velocity are calculated in real time in the collecting process; and in the collecting process, the filter membranes are cooled in real time, and the cooling temperature is 3-5 DEG C.
