Abstract

The invention provides a screening model and screening method for anticancer drugs targeting a LINE-1 ORF1 protein. The drug screening model is breast cancer cell T47D with high expression of LINE-1 ORF1. An In-Cell Western method is adopted to detect an expression quantity of the LINE-1 ORF1 protein in cells after a compound is added, and a simple, sensitive and stable method for quantitatively analyzing the expression quantity of the LINE-1 ORF1 protein is established, thereby verifying the possibility that the screening model is used as a high-throughput screening system for LINE-1 ORF1 protein inhibitors and agonists. A plurality of inhibitors targeting the tumor cell LINE-1 ORF1 protein have been successfully obtained by using the screening model, and a new means is provided for the development of anti-tumor drugs.

Description

technical field [0001] The invention relates to the field of biomedicine, in particular to an anticancer drug screening model and screening method with LINE-1 ORF1 protein as the target. Background technique [0002] In the 1940s, American scientist Barbara McClintock discovered a genetic component that can move freely within the chromosome or between different chromosomes independently of the cell mitosis process when studying the cause of variegated corn. This genetic component was later named transposition Son (transposon), also known as transposable elements (transposable elements, TEs). [0003] According to the different transposition methods, transposons can be divided into two categories: DNA transposons and RNA transposons. A DNA transposon is a DNA segment encoded with a transposase, which can cut its own DNA segment from its original genomic position and insert it into other positions in the genome. This replication method is vividly described as "Cut-paste" mod...
