Abstract
The invention provides a screening model and screening method for anticancer drugs targeting a LINE-1 ORF1 protein. The drug screening model is breast cancer cell T47D with high expression of LINE-1 ORF1. An In-Cell Western method is adopted to detect an expression quantity of the LINE-1 ORF1 protein in cells after a compound is added, and a simple, sensitive and stable method for quantitatively analyzing the expression quantity of the LINE-1 ORF1 protein is established, thereby verifying the possibility that the screening model is used as a high-throughput screening system for LINE-1 ORF1 protein inhibitors and agonists. A plurality of inhibitors targeting the tumor cell LINE-1 ORF1 protein have been successfully obtained by using the screening model, and a new means is provided for the development of anti-tumor drugs.
