Problems solved by technology

However, since key factors such as hormones and proteins related to female reproduction are greatly affected by the menstrual cycle, the sampling time of serum samples is difficult to be uniform, and it is greatly affected by other systems in the body, which brings challenges for the subsequent verification of large samples. difficulty

Embodiment Construction

[0031] The inventors collected patients with unexplained recurrent spontaneous abortion (URSA) who did not obtain normal clinical pregnancy in this egg retrieval cycle, and patients with tubular factor infertility who obtained live birth in this egg retrieval cycle as normal controls (Healthy control), through TMT (Tandem masstag) technology, the determination of differentially expressed proteins at the proteomics level in follicular fluid. Through the preliminary screening of differentially expressed proteins and the ELISA verification of further expanded samples, it was found that HRG was expressed at a low level and C4B was expressed at a high level in URSA patients. Further ROC (receiver operator characteristic curve) analysis showed that the ROC curve was drawn with the concentration of HRG follicular fluid as the test variable and whether the patient obtained a live birth in this egg retrieval cycle as the state variable. The area under the curve (AUC) ) was 0.785 (P<0.0...

Abstract

The invention belongs to the field of reproductive medicine. According to the technical scheme provided by the invention, the follicular fluid HRG and C4B protein expression level is used as an early warning method for unexplained recurrent abortion. The method is not used for disease diagnosis and treatment, and comprises the following steps of 1) collection and preservation of follicular fluid: picking up an ovum crown compound from the collected follicular fluid at a constant temperature, then collecting the remaining follicular fluid, centrifuging at 1500 rpm to take a supernatant, subpackaging, and cryopreserving at -80 DEG C, (2) carrying out ELISA determination on the protein concentration of C4B and HRG follicular fluid: carrying out corresponding protein concentration detection on the follicular fluid by adopting an ELISA detection kit of corresponding protein, and 3) result statistics and analysis. The method has a certain prediction value for the pregnancy outcome of the URSA patient.

Description

technical field [0001] The invention belongs to the field of reproductive medicine, in particular to an early warning method for unexplained recurrent miscarriage patients. Background technique [0002] Among couples of childbearing age, the rate of miscarriage in a single pregnancy is approximately 15-25%, and most miscarriages occur in the first trimester. About 2-5% of couples of childbearing age will experience 2 or more miscarriages. Three or more fetal losses before 28 weeks of gestation are usually referred to as recurrent miscarriage. The etiology of recurrent miscarriage is complex. Common causes of recurrent miscarriage in the first trimester include genetic factors, anatomical abnormalities, infectious factors, and autoimmunity. Diseases, endocrine abnormalities, prothrombotic state and environmental factors, etc. However, more than half of RSA cases cannot be definitively diagnosed with etiology, which is called unexplained recurrent spontaneous abortion (URSA)...
