Embodiment 1

[0043] Example 1 Construction of CECT-based radiomics score as a predictor of lesion responsiveness in hepatocellular carcinoma receiving anti-PD-1 therapy

[0044] This study collected HCC patients who received anti-PD-1 therapy at Nanfang Hospital of Southern Medical University between August 2018 and October 2020. This study was approved by the Review Board of Nanfang Hospital, Southern Medical University and complies with the Declaration of Helsinki.

[0045] The inclusion criteria were as follows: (a) diagnosed as HCC after biopsy or according to AASLD treatment guidelines; (b) receiving anti-PD-1 therapy.

[0046] Patients with the following characteristics were excluded: (a) contrast-enhanced CT (contrast enhanced CT, CECT) was not available at baseline examination; (b) treatment duration was <6 weeks; (c) imaging artifacts were present; (d) presence of History of other malignancies; (e) no measurable target lesions at baseline.

[0047] A measurable target lesion was...

Abstract

The invention discloses a PD-1 curative effect prediction model based on radiomics and a construction method thereof. The construction method of the model comprises the following steps: collecting image data of a liver cancer patient, extracting radiomics characteristics, screening key characteristics by using LASSO logistic regression, and carrying out weighted construction through a regression coefficient to obtain a corresponding PD-1 curative effect prediction model based on radiomics. According to the method, a lesion-based method is adopted, extracted features are evaluated and predicted on the lesion level, it is found through research that a radiomics score may be a prediction factor of lesion reactivity of hepatocellular carcinoma patients subjected to anti-PD-1 treatment, and an average radiomics score may have the ability of screening inferior-effect patients subjected to anti-PD-1 treatment; and a certain clinical value is achieved.

Description

technical field [0001] The invention belongs to the technical field of precision medicine, and in particular relates to a radiomics-based PD-1 curative effect prediction model and a construction method thereof. Background technique [0002] Hepatocellular carcinoma (HCC) is the fourth most common cause of cancer-related death worldwide and is a major global health problem. The emergence of immunotherapy has greatly changed the treatment strategy of HCC. But not all patients benefit from immunotherapy. Coupled with the high cost of immunotherapy and the possible serious adverse reactions, it is necessary to effectively stratify patients according to the therapeutic effect before receiving immunotherapy. [0003] Existing studies have used radiomics to predict the response of HCC to immunotherapy. Yuan et al. established a radiomics nomogram to evaluate the effect of anti-PD-1 therapy in patients with HCC. However, they perform assessment, feature extraction and prediction...
