Problems solved by technology

There are many disadvantages in the method of dividing into balls: 1. Low reproduction rate: unable to meet the needs of the market
Therefore, using these two methods to breed Fragrant Snowland can not meet the rapidly growing market demand

Embodiment Construction

[0019] The best embodiment of the present invention will be described in detail below by way of examples.

[0020] 1. Induction of callus or direct somatic embryos:

[0021] The explants were surface-sterilized with 70% alcohol under aseptic conditions, and then soaked in 0.1% mercury liter for 3-5 minutes. After being washed several times with sterile water, the explants were cut into small pieces of a certain length [young spike (1-2 mm), inflorescence rachis (2-3 mm), stem (1-3 mm), leaf (2 -3 mm), flower base (3-4 mm), mature embryo (1-1.5 mm), immature embryo (0.5-1 mm), root (1-2 mm)]. Small pieces of explants can be inoculated on the following two induction media to induce the formation of somatic embryos or callus.

[0022] Induction medium I (for direct induction of somatic embryos): N 6 +1~2mg / L 2,4-dichlorophenoxyacetic acid (2,4-D)+2~4mg / L 6-benzylaminopurine (6-BA) solid agar medium;

[0023] Induction medium II (for callus induction): solid agar medium of MS+...

Abstract

The invention belongs to flower cultivation and reproduction technology, and in particular relates to in vitro tissue culture and soilless bulb breeding technology of modern polyploid freesia x hybrida. The invention discloses a method for in vitro rapid plant regeneration and soilless bulb propagation of modern polyploid Cymbidium syringae by applying techniques such as in vitro tissue culture and low temperature treatment. Under in vitro culture conditions, young spikes, inflorescence rachis, stems, leaves, flower bases, mature embryos, immature embryos, roots, etc. are used as explants to induce plant regeneration by modulating the medium. After special treatment, the soilless bulb propagation of Fragrant Snowland has been realized. The invention also discloses a cryopreservation method for the callus of the sweetsnow plant. The invention has the advantages of: 1. short breeding cycle and high reproduction coefficient; 2. non-toxic bulbs; 3. land saving; 4. production is not limited by seasons and the like. The present invention can be applied to the field of large-scale production of the flowers.

Description

technical field [0001] The invention belongs to flower cultivation and propagation technology, in particular to in vitro tissue culture and soilless bulb propagation technology of modern polyploid Freesia x hybrida. Background technique: [0002] Freesia is also known as freesia, calamus, fragrant iris, and tuberose. The classification of freesia is a perennial bulbous herbaceous flower belonging to the genus of the family Iridaceae. [0003] The modern polyploid Snowberry is cultivated from the wild diploid Snowberry by artificial breeding technology, and its somatic cell chromosome number is 33 or 44. Modern polyploid Snowland is a rare cut flower variety, which has the advantages of long flowering period, tall plant, large flower, bright color, fragrant flower and long flower arrangement period. Another feature of Fragrant Snowland is that its flowering period is in winter and spring, and it can be supplied to the market in early spring. It can be cultivated as a cut f...
