Problems solved by technology

[0004] In the United States, people have carried out research on the separation and extraction of type II collagen using one of the jellyfish, Stomolophus meleagris, as a raw material, and have applied for a patent [1]. The research on type II collagen has not been reported at home and abroad

[0012] Take 1000g of fresh frozen jellyfish, thaw it, wash it, and process it with a high-speed tissue homogenizer at 10000rpm for 10 minutes at room temperature to obtain a homogenate. Add 2 volumes of 0.1 mol / L NaOH to the homogenate, stir at 4°C for 24 hours, then centrifuge at 8,000 rpm for 10 min at 4°C in a high-speed refrigerated centrifuge, discard the supernatant, and collect the precipitate. The above-mentioned extraction operation on the homogenate was repeated 3 times. Rinse the precipitate after centrifugation repeatedly with distilled water until neutral, then add 0.5mol / L acetic acid, stir at 4°C for 48 hours, then centrifuge at 4°C, 8000rpm for 20min with a refrigerated centrifuge, discard the supernatant , to collect the precipitate. Repeat the above extraction operation 2 times. Add 2 times the volume of 0.5 mol / L acetic acid to the precipitate obtained by centrifugation, add pepsin at 5‰ weight of jellyfish, stir for at least 48 hours, then use a refrigera...

[0014] Take 1000g of commercially available hemp stingers marinated with three alum, drain the water, soak in 2-5 times the amount of deionized water at room temperature, and change the water every 2-3 hours until the content of sodium chloride in the soaking water is low Up to 1.5ppm (chloride ion test was negative), 980g of desalted jellyfish were obtained. The desalted jellyfish was treated with a high-speed tissue homogenizer at 10,000 rpm for 10 minutes at room temperature to obtain a homogenate. Add 2 times the volume of 0.1 mol / L NaOH to the homogenate, stir at 4°C for 24 hours, then centrifuge at 4°C, 8000rpm for 30min in a high-speed refrigerated centrifuge, discard the supernatant, and collect the precipitate. The above-mentioned extraction operation on the homogenate was repeated 3 times. Rinse the precipitate after centrifugation repeatedly with distilled water until neutral, then add 0.5mol / L acetic acid, stir at 4°C for 48 hours, then centrifuge at 4°C, 8000rpm ...

example 3

[0016] A liquid sample of type II collagen was assayed for protein content, total sugar content, and amino acid composition. The measurement results showed that the sample contained 98% protein and basically no sugar. The composition of the amino acids contained in it is shown in Table 1.

[0017] The amino acid composition of table 1 test sample

[0018] amino acid

[0019] arginine

Abstract

The present invention provides type II collagen from cyanea arctica and the preparation process, pertaining to the natural biological active material and the technical art of preparation. The present invention provides novel natural halobios type II collagen. Being operated according to the present invention, 0.5 percent wt. type II collagen with purity up to more than 85 percents can be obtained. The type II collagen is originally white powder with fishy smell, which contains 85 to 99 percents wt. protein and almost no sugar. The type II collagen is nontoxic and has the reactivity of II collagen such as oral medication, inducement, immunological tolerance, prevention and treatment of chronic infectious arthritis.

Description

technical field [0001] The patent relates to type II collagen derived from jellyfish and its preparation process, which belongs to the technical field of natural bioactive substances and its preparation. Background technique [0002] Cyanea nozakii, commonly known as hemp sting, is a large marine plankton belonging to Phylum Cnidaria, Class Scyphomedusae, Order Phylum cnidaria, and family Jellyfish. (Family Cyanea). There are four species of white jellyfish (Cyanea nozaki Kishinouye), hair-shaped jellyfish (Cyanea capillata), brown jellyfish (Cyanea ferruginea Eschscholtz), and purple jellyfish (Cyanea purpurea Kishinouye) that have been found along the coast of my country. The most and the widest distribution. The jellyfish usually uses small zooplankton as bait. It has well-developed gonads, strong reproductive ability, and extremely fast growth. At present, the annual output of jellyfish in my country has reached 10 million tons, forming a new natural marine biological r...
