Problems solved by technology

Scholars have used different molecular marker techniques (including mitochondrial control region, AFLP, RAPD, etc.) to study the genetic diversity of Chinese alligator, but the above genetic markers cannot provide sufficient genetic information, such as Wang et al., 2003: D-loop sequence variation of mitochondrial DNA in captive Chinese alligator. Acta Genetic Sinica, 30, 425-430; Wu et al., 2002: Genetic variation in captive population of Chinese alligator, Alligator sinensis, revealed by random amplified polymorphic DNA (RAPD). Biological Conservation3, 106, 441; Yan et al., 2006: AFLP analysis of genetic variation on captive-bred Chinese alligators: an application to select individuals for release. Zoo Biology , 25(6), 479-490

Embodiment Construction

[0020] The present invention will be described in further detail below in conjunction with the accompanying drawings.

[0021] 1: Construction of Chinese alligator microsatellite DNA enrichment library:

[0022] a) Digestion and ligation of genomic DNA: Alligator genomic DNA was extracted according to the method of Sambrook et al. (2002, Molecular Cloning, 3nd 463-471), and digested with restriction endonuclease Bsp 143I (purchased from Fermentas). The digested product was added to adapters SauLA (5'-GGCCAGAGACCCCCAAGCTTCG-3'), SauLB (5'-GATCCGAAGCTTGGGGTCTCTGGCC-3'), 10×ligation Buffer, T4 DNA ligase (purchased from Promega) and ligated overnight at 16°C in a PCR machine.

[0023] b) Using SauLA as a primer and the above ligation product as a template, carry out a PCR reaction, electrophoresis on a 2.0% agarose gel, cut out <1kb fragments under ultraviolet light, and use a gel recovery kit (purchased from Axygen) to carry out Recycle.

[0024] c) Using the recovered DNA as ...

Abstract

The invention discloses an alligator sinensis microsatellite DNA mark which includes the construction of the enriched libraries of (CT / AG)n, (AC / GT)n and (GATA)n of an alligator sinensis microsatellite, the screening and sequencing of the positive clones of a microsatellite sequence, obtaining 25 clones which comprise the repeated sequence of the microsatellite and conforming 10 microsatellite marks of Asiu1, Asiu2, Asiu4, Asiu20, Asiu44, Asiu45, Asiu84, Asiu96, Asiu122 and Asiu234 of abundant polymorphism. The alligator sinensis microsatellite DNA mark can be used for the studies on genetic diversity of different geographic populations of the alligator sinensis, has good repetitiveness and is a reliable and effective molecule mark.

Description

technical field [0001] The invention relates to a molecular marker technology, in particular to a Chinese alligator microsatellite DNA genetic marker. Background technique [0002] Microsatellite DNA, also known as short tandem repeats (STR), simple repeats (SSR), simple sequence length polymorphism (SSLP), refers to the tandem repeats of 1-6 nucleotides in the genome. Nucleotide sequence; among them, the most common dinucleotide repeats are (AC / TG)n, (AG / TC)n. Different organisms show polymorphism due to the different number of repetitions of the repeating unit. Studies have shown that the "strand slippage" phenomenon in the DNA replication process may cause the high polymorphic information content (PIC) of microsatellite DNA. The high mutation, co-dominant expression and ubiquity of microsatellite DNA in eukaryotic genomes make it a superior molecular marker for population genetics research, identification of germplasm resources, kinship analysis and map construction.
