Problems solved by technology

Therefore, the establishment of a stable tissue culture regeneration system for Lianhong is the basis for genetic transformation of Lianhong. Currently, regenerated plants are obtained by inducing callus and differentiation of explants such as stems, leaves, and root tips. The probability is quite low, just accidental, and sometimes even no differentiated seedlings can be obtained (Yang Xiaohong et al., 2007, December, Agricultural Science and Technology Communication), so it is difficult to meet the genetic transformation requirements of exogenous genes

Embodiment 2

[0057] Embodiment 2, domestication and transplanting of regenerated plants:

[0058] The regenerated plants obtained in Example 1 (that is, the seedlings planted out of the bottle) together with the culture bottle were taken out of the culture room and placed in the greenhouse for seedling hardening. At this time, the bottle cap was not opened, and the bottle mouth was opened after 1 week, and a small amount of Water (the amount of water added is generally controlled so that there is a layer of water on the medium). The seedlings are hardened and transplanted after 2 to 3 days. The environment in the greenhouse is: 25±2°C, natural light.

[0059] A string of red regenerated seedlings was transplanted in the greenhouse, the root medium was washed off during transplanting, and the seedlings were transplanted into a nutrient pot (the soil quality was nutrient soil and perlite in a weight ratio of 8:1). When the seedlings grow to a height of more than 5 cm, they are transplanted i...

Abstract

The invention discloses a method for regenerating salvia splendens Ker-Gawl plant, comprising the following steps in sequence: 1) sterilizing a seed of salvia splendens Ker-Gawl; 2) aseptically culturing the seed of salvia splendens Ker-Gawl; 3) longitudinally cutting the seedling after aseptic culture into two parts, removing peel before inoculating the seedling in a callus induced culture medium with the cutting surface facing downwards; 4) inoculating the induced product in step 3) on the callus induced culture medium for subculture 1-2 times; 5) transplanting the resultant well-grown callus after subculture on a differential medium to carry out adventitious bud differentiation; 6) cutting the adventitious bud in step 5) as a small seedling to be transplanted on a rooting medium for rooting culture till the small seedling has at least three adventitious roots which have the length not less than 2 cm; and obtaining seedling which can be transplanted out of the bottle, wherein the steps 1) to 6) are performed in aseptic environment. Through the application of the method, a great deal of regenerative salvia splendens Ker-Gawl plant can be obtained.

Description

technical field [0001] The invention belongs to the field of agriculture, and in particular relates to a method for obtaining regenerated plants from calluses. Background technique [0002] A bunch of red (Salvia splendens Ker-Gawl) belongs to the Labiatae family (Labiatae), and is a perennial herb of the genus Salvia (Salvia L). It is native to the tropical regions of South America. Now it is widely cultivated in various parts of China, often as an annual cultivation. A string of reds is widely used to decorate city squares, flower beds, flower borders, roads, courtyards, etc. due to its short growth cycle, short and sturdy plants, strong adaptability, and bright colors. Moreover, the red color of its calyx and corolla is It is inferior to other flowers and plants, and should be arranged with light-colored flowers. It is the most commonly used red grass flower to decorate the environment and set off the festive atmosphere in Chinese festivals (Li Fenglan et al., Journal of ...
