Embodiment 1

[0059] Example 1 Detection of GADD45A gene mutation and its association with ovarian cancer

[0060] 1.1 Research object

[0061] A total of 139 patients with ovarian cancer were selected and diagnosed and treated in Qilu Hospital of Shandong University from September 2008 to April 2011. The clinical data were obtained from medical records. 189 age-matched control patients were healthy women who underwent physical examination in the Health Examination Center of Qilu Hospital. Most of the subjects were Han residents in Shandong Province. Take 1.5ml of peripheral blood from the test personnel and store it in a freezer at -80°C. All subjects obtained consent in accordance with the requirements of the Ethics Committee of Qilu Hospital of Shandong University.

[0062] 1.2 DNA extraction

[0063] The peripheral blood DNA of the subjects was extracted by the conventional phenol-chloroform method, as follows:

[0064] (1) Take 400ul of blood and add it to a 1.5ml centrifuge tube....

Embodiment 2

[0087] Example 2 Ovarian Cancer Susceptibility Detection Kit

[0088] Since the 1481G>A mutation in SEQ ID NO: 1 is highly correlated with ovarian cancer publications, GADD45A gene-specific primers can be designed based on this mutation and then expanded detection using the patient's DNA as a template.

[0089] Prepare a kit (100 person-times), which contains the substances shown in Table 3:

[0090] table 3

[0091]

[0092] 2ml of peripheral blood was drawn from the subjects, and DNA was extracted by conventional methods. Ovarian cancer susceptibility detection kit was used for PCR reaction, the reaction products were sequenced, and the sequencing results were checked and analyzed using Meglign 7.0 and Chromas 2.33 software. The susceptibility of subjects with rs2759219 (1481G>A) in the test results to ovarian cancer was lower than that of the normal population.

Abstract

The invention discloses a molecular marker for identifying the susceptibility of an ovarian cancer. The nucleotide sequence of the molecular marker is shown as SEQ ID NO:1, and a single nucleotide polymorphism G)A exists at a bit 1481. A kit for identifying the susceptibility of the ovarian cancer comprises a pair of specific primers of which the sequences are shown as SEQ ID NO:2 and SEQ ID NO:3. A method for identifying whether the single nucleotide polymorphism of a GADD45A gene exists in a sample in vitro comprises the following steps of: (1) amplifying the GADD45A gene of the sample by using specific primers to obtain an amplification product; and (2) sequencing the amplification product and detecting whether a single nucleotide polymorphism, i.e., 1481G)A exists in the amplification product. The method is easy, accurate and rapid for identifying, has high specificity, and has great significances on effective early-diagnosis, early-treatment and personal prevention and treatment on high-risk groups or susceptible individuals and patients suffering from the ovarian cancers.

Description

technical field [0001] The present invention relates to a molecular marker, kit and identification method for identifying the susceptibility of ovarian cancer, in particular to the single nucleotide polymorphism (single nucleotide polymorphism, SNP) of GADD45A gene and its correlation with ovarian cancer Properties, and methods and kits for identifying these SNPs belong to the fields of molecular biology and medicine. Background technique [0002] Ovarian cancer is the most lethal tumor of the female reproductive system. In 2008, there were 225,500 new cases and 140,200 deaths worldwide. Due to insidious onset and inconspicuous symptoms, most patients are diagnosed at an advanced stage. The 5-year survival rate of patients with advanced ovarian cancer is only about 30-45%, and the prognosis is poor. However, the 5-year survival rate of patients with early ovarian cancer is as high as 90%. It reminds us that the key to improving the prognosis and survival of ovarian cancer p...
