Abstract

The invention discloses a technical method for promoting the tissue culture rooting of alpine rose amethyst. The method comprises the following steps of: cutting off robust sterile cluster buds by scissors on a sterile ultra-clean work table, and inoculating into the following rooting culture medium, namely a 1/2 WPM minimal medium, adding 10mg/L of added NAA (l-naphthylacetic acid), 10g/L of cane sugar, 0.3g/L of activated carbon and 6g/L of agar powder. The method comprises the following steps of: firstly, carrying out 3 days of dark culture, carrying out 7 days of light culture, taking out bud seedlings on the sterile ultra-clean work table, and transferring into the hormone-free 1/2 WPM minimal medium to continuously carry out 30-40 days of light culture. After the method is used, the rooting percentage of the tissue culture seedling can be improved by more than 30%, the rooting percentage of the amethyst tissue culture seedling can be greatly improved, the tissue culture seedling root system treated by the method has thicker and more secondary branches and can be more easily transplanted to be survived.
