Embodiment Construction

[0012] The present invention will be described in detail below in conjunction with specific embodiments.

[0013] 1. DNA extraction

[0014] Use muscle or blood tissue to extract the genomic DNA of Chen's new whitebait. Genomic DNA was extracted using SDS / proteinase K cleavage and phenol-chloroform extraction (Sambrook et al., 1999), see figure 1 .

[0015] 2. Screening of Microsatellite Enrichment Library of Chen's New Silverfish

[0016] Restriction ligation Genomic DNA was extracted, partially digested with the restriction endonuclease Sau3AI, and then ligated with the Sau3AI linker (A) 5'-GATCGTCGACGGTACCGAATTCT / (B) 3'-CAGCTGCCATGGCTTAAGA ACTG.

Abstract

The invention discloses neosalanx tangkahkeii sequence tagged microsatellite sites, primers and application of the neosalanx tangkahkeii sequence tagged microsatellite sites and primers. The sequence tagged microsatellite sites comprise 12 stable polymorphism sequence tagged microsatellite sites of which the sequences are respectively shown in SEQ ID NO:1-SEQ ID NO:12. Sequences of primers of the neosalanx tangkahkeii sequence tagged microsatellite sites are respectively shown in SEQ ID NO:13-SEQ ID NO:36. The invention also discloses 12 microsatellite sequences and 12 pairs of microsatellite primers of neosalanx tangkahkeii. The sites have significances in conservation genetics research, germplasm resource investigation and assistant breeding of neosalanx tangkahkeii.

Description

technical field [0001] The invention relates to the field of biotechnology, in particular to the microsatellite site, primer and application of Chen's new whitebait. Background technique [0002] Neosalanx tangkahkeii (Neosalanx tangkahkeii) belongs to the genus Neosalanx of the order Smeltfish, also known as Taihu Newbait, near Taihu Lake Neosalanx, it is a unique species in China, distributed in the middle and lower reaches of the Yangtze River, including its tributaries and subsidiary lake. Chen's new whitebait is a traditional economic fish in my country, especially freshwater fish. However, due to water pollution, overfishing, and habitat changes, the number of Newbait Chenshi has been greatly reduced, so the protection of Newbait Chenshi is urgent. In recent years, microsatellite markers have played an important role in the study of population structure. Contents of the invention [0003] The technical problem to be solved by the present invention is to provide Ch...
