[0027] 1.1 Materials

[0028] 1.1.1 From April 2015 to August 2015, 7 cases of DDH patients and 6 cases of femoral neck fracture patients were collected from Xinhua Hospital Affiliated to Shanghai Jiaotong University, including 8 males and 5 females, age 31-76 years old, with an average of 51.1±14.4 years old. This study was approved by the Hospital Medical Ethics Committee and the patients gave informed consent.

Abstract

The invention relates to a novel application of Lnc RNA H19 (long non-coding ribonucleic acid H19), that is, overexpression of Lnc RNA H19 can delay the progression of disease from DDH (developmental dysplasia of hip) to osteoarthritis. The application has the advantages that the research of 7 cases of DDH cartilage tissue and 6 cases of cartilage tissue of fractured femoral neck shows that the expression of Lnc RNA H19 in DDH is remarkably reduced compared with that of Lnc RNA H19 in femoral neck fracture, and H19 is possible to have a relation with occurrence and development of DDH; DDH is accelerated to deteriorate into osteoarthritis if H19 is knocked down; the expression of H19 can be promoted through transfection of a slow virus LV-H19, and the overexpression of H19 can delay the progression of disease from DDH to osteoarthritis.

Description

technical field [0001] The invention relates to the technical field of molecular biology, in particular to the application of long-chain non-coding RNAH19 (LncRNAH19) in developmental dysplasia of the hip (DDH). Background technique [0002] Long non-coding RNA (long non-coding RNA, lncRNA) is defined as a class of RNA transcripts longer than 200 nt that do not encode proteins. It was originally considered to be the "noise" of genome transcription, just a by-product of RNA polymerase II transcription. Has biological functions. However, studies in recent years have shown that lncRNAs are involved in many important regulatory functions such as X chromosome silencing, genome imprinting and chromatin modification, transcriptional activation, transcriptional interference, nuclear transport, protein post-translational modification, and epigenetic regulation of genetics. process. These effects indicate that it is closely related to the pathophysiological changes of diseases, espe...
