The invention relates to the field of fertilizer processing and provides a preparation method of a bio-organic fertilizer. The preparation method comprises the following specific steps: uniformly mixing the following base materials (in parts by weight): 70-95 parts of animal dung and 5-30 parts of plant straw; sprinkling a compound bacterial agent, controlling the water content to 50-55%, and fermenting for 6-9 days, wherein 1-2kg of MP compound probiotics and 1-2kg of MP partner are added into each ton of base materials, and the MP compound probiotics comprise 7-12 parts of compound strains, 7-12 parts of syrup, 1-1.5 parts of bran, 0.05-0.1 part of carbamide, 0.05-0.1 part of NaCl, 0.01-0.05 part of tea and 180-250 parts of water; and performing secondary turning: throwing and turning the obtained fermented material, controlling the water content to 40-45%, and fermenting for 6-9 days. The bio-organic fertilizer prepared by the preparation method has the advantages of high fermentation efficiency, short period and high composting degree.
