Applications of gancaonin M in preparation of medicines treating and / or preventing thrombotic diseases are disclosed. Experiments prove that the gancaonin M can significantly reduce the weight of vein bypass thrombi in vivo and significantly increase NO, cGMP and PGI2 levels in plasma of rats affected with thrombi; the gancaonin M can inhibit platelet aggregation through influencing a NO / cGMP metabolic pathway and changing a PGI2 level; the gancaonin M can significantly relieve the degree of encephaledema caused by cerebral thrombosis, can reduce the MDA content in brain tissue and can increase SOD activity; and the gancaonin M has certain anti-oxidation activity and has an obvious protective effect on cerebral thrombi.
